Biorhizome: A Biosynthetic Platform for Colchicine Biomanufacturing

Abstract: Colchicine is one of the oldest plant-based medicines used to treat gout and one of the most important alkaloid-based antimitotic drugs with anticancer potential, which is commercially extracted from Gloriosa superba. Clinical trials suggest that colchicine medication could prevent atrial fibrillation recurrence after cardiac surgery. In addition, therapeutic colchicine is undergoing clinical trials to treat non-diabetic metabolic syndrome and diabetic nephropathy. However, the industrial-scale biomanufacturin… Show more

“…In this species (C. hierosolymitanum), the maximum yield of colchicine was also noted in callus added to 30 mM NH 4 and 0.1 M of sucrose (30). Sivakumar et al (31) described the importance of biorhizomes, rhizome specific genes/gene which over expressed in rhizomes for the biomanufacture of colchicine as root, callus and cell suspension of G. superba and various species of Colchicum yielded poor level of colchicine. It is believed that the synthesis of colchicine and colchicoside, a related secondary metabolite are often controlled by various external factors; our results thus indicate that NaCl could be a good inducer in improving colchicine yield, therefore of huge practical applications.…”

“…The Gloriosa biorhizoma produces compound 1 constantly regardless of the weather or the seasons of the year. However, for a more cost-effective production of compound 1 by this new technology and its massive use, it is necessary to understand the molecular mechanisms in the biorhizoma and the biosynthetic pathway of compound 1 in more detail and to use molecular engineering methods to start overproduction of this alkaloid in the bioreactor [ 42 ].…”

“…So far, this method has used only CLC to polyploidise daylilies. CLC is highly toxic to animal cells, has anticancer effects and is used in chemotherapy to prevent the growth of cancer cells in medicine (Sivakumar et al, 2017). ORZ is used as an alternative to CLC -it is less toxic to animals than CLC.…”
