Bioreducible Hyperbranched Poly(amido amine)s for Gene Delivery

Abstract: A series of reducible hyperbranched poly(amido amine)s (RHB) with high transfection efficiency were designed and synthesized as non-viral gene delivery vectors. The polycations were synthesized by Michael addition copolymerization of N,N-dimethylaminodipropylenetriamine (DMDPTA) and two bisacrylamide monomers N,N′-hexamethylene bisacrylamide (HMBA) and N,N′-cystamine bisacrylamide (CBA). The density of disulfide linkages in the synthesized hyperbranched polymers was tuned by varying the feed molar ratio of the… Show more

“…Therefore Oupicky et al prepared a complete series of reducible hyperbranched PAMAMs (RHB), which were synthesized by Michael addition copolymerization of N,Ndimethylaminodipropylene-triamine (DMDPTA) with either N,N 0 -hexamethylene bisacrylamide (HMBA) or N,N 0 -cystamine bisacrylamide (CBA) (Fig. 10) [118]. A library of compounds with different co-monomer ratios was built, with special emphasis given to the preparation of RHB with different disulfide loadings.…”

“…Furthermore, bioreducibility of transfection agents is essential for the release of the complexed nucleotides inside the cells with subsequent elimination of the degradation products [10]. Amongst several transfection agents explored in clinical studies so far [11,12], the supramolecular cationic cyclodextrin polymers have also yielded appreciable results [13].…”

“…Vectors such as cationic lipids and polymers have been widely studied [21][22][23]. Among those, cationic polymers, such as poly(L-lysine) (PLL) and polyethylenimine (PEI), are the best known [24].…”
