Wawekale Rainforest Reserve (WRR) is a primary lowland tropical rainforest area in the Western Province of Sri Lanka, established with diversity, but disturbed by human activities such as rubber cultivation, logging, poaching, chemical release, and the collection of firewood. Visual Encounter Surveys (VES) were conducted for 100 hours in the year 2022, which covered wide habitat distribution. The herpetofaunal diversity was measured in the WRR which can indicate broader ecological shifts, environmental changes, or habitat degradation. Water quality was measured in the WRR area to determine the ecosystem health and human impact. The study reports 171 species in 73 families, including two Critically Endangered (1.17%), six Endangered (3.51%), 16 Vulnerable (9.36%), and 14 Near Threatened (8.19%) species according to the IUCN Red List. The study area showed 51 endemic faunal species in Sri Lanka, which represented 29.83% of endemism. Based on the observations conducted on the herpetofauna diversity, the Shannon-Wiener diversity index (H) was calculated and the overall herpetofauna (H’H) showed high diversity within the forest premises. The water quality of the forest fragment had an influence from anthropogenic activities such as removing forest cover, bathing, washing vehicles, and releasing agro-chemical compounds resulting in high water temperature, basic pH level, high electrical conductivity, and high total dissolved solid conditions. The WRR, designated under protection, exhibits the need for immediate remedial actions to mitigate human-induced pollution and deforestation, emphasizing the critical importance of implementing proactive conservation measures to sustain ecological integrity and preserve biodiversity.

Background Tilapia lake virus (TiLV), also known as Tilapinevirus tilapiae, poses a significant threat to tilapia aquaculture, causing extensive mortality and economic losses. Understanding the mechanisms and pathogenesis of TiLV is crucial to mitigate its impact on this valuable fish species. Methodology In this study, we utilized transmission electron microscopy to investigate the ultrastructural changes in E-11 cells following TiLV infection. We also examined the presence of TiLV particles within the cells. Cellular viability and mitochondrial functions were assessed using MTT and ATP measurement assays and mitochondrial probes including JC-1 staining and MitoTracker™ Red. Results Our findings provide novel evidence demonstrating that TiLV causes cytotoxicity through the destruction of mitochondria. Transmission electron micrographs showed that TiLV particles were present in the cytoplasm of E-11 cells as early as 1 h after infection. Progressive swelling of mitochondria and ultrastructural damage to the cells were observed at 1, 3 and 6 days post-infection. Furthermore, losses of mitochondrial mass and membrane potential (MMP) were detected at 1 day after TiLV inoculation, as determined by mitochondrial probes. The results of the MTT assay also supported the hypothesis that the cell deaths in E-11 cells during TiLV infection may be caused by the disruption of mitochondrial structure and function. Conclusions Our study reveals the significant role of mitochondrial disruption in contributing to cellular death during the early stages of TiLV infection. These findings advance the understanding of TiLV pathogenesis and further enhance our knowledge of viral diseases in fish.

Nile tilapias are known to be an economically important farmed fish in Indonesia. Due to its adaptability, this tilapia fish can thrive in various aquatic conditions, making them an important commercial fish to produce locally or export internationally. Because of this potential, this study aims to provide recommendations using the decision support system to facilitate fish farmer decision-making with the Simple Additive Weighting model. This approach is expected to provide recommendations to fish farmers, or the general public interested in having their own Nile tilapia brood stock. The fish in this research was selected randomly following six criteria, namely fish weight, length, movement, fin or body deformation, fish colour and fish stomach condition. The result-based decision making model in this study provides an accuracy of 86.6%.
