Microalgae have great prospects as a sustainable resource of lipids for refinement into nutraceuticals and biodiesel, which increases the need for detailed insights into their intracellular lipid synthesis/storage mechanisms. As an alternative strategy to solvent-and label-based lipid quantification techniques, we introduce time-gated coherent anti-Stokes Raman scattering (CARS) microscopy for monitoring lipid contents in living algae, despite strong autofluorescence from the chloroplasts, at approximately picogram and subcellular levels by probing inherent molecular vibrations. Intracellular lipid droplet synthesis was followed in Phaeodactylum tricornutum algae grown under (1) light/nutrient-replete (control [Ctrl]), (2) light-limited (LL), and (3) nitrogenstarved (NS) conditions. Good correlation (r 2 = 0.924) was found between lipid volume data yielded by CARS microscopy and total fatty acid content obtained from gas chromatography-mass spectrometry analysis. In Ctrl and LL cells, micron-sized lipid droplets were found to increase in number throughout the growth phases, particularly in the stationary phase. During more excessive lipid accumulation, as observed in NS cells, promising commercial harvest as biofuels and nutritional lipids, several micron-sized droplets were present already initially during cultivation, which then fused into a single giant droplet toward stationary phase alongside with new droplets emerging. CARS microspectroscopy further indicated lower lipid fluidity in NS cells than in Ctrl and LL cells, potentially due to higher fatty acid saturation. This agreed with the fatty acid profiles gathered by gas chromatography-mass spectrometry. CARS microscopy could thus provide quantitative and semiqualitative data at the singlecell level along with important insights into lipid-accumulating mechanisms, here revealing two different modes for normal and excessive lipid accumulation.

Most microorganisms remain uncultivated, and typically their ecological roles must be inferred from diversity and genomic studies. To directly measure functional roles of uncultivated microbes, we developed Chip-stable isotope probing (SIP), a high-sensitivity, high-throughput SIP method performed on a phylogenetic microarray (chip). This approach consists of microbial community incubations with isotopically labeled substrates, hybridization of the extracted community rRNA to a microarray and measurement of isotope incorporation-and therefore substrate use-by secondary ion mass spectrometer imaging (NanoSIMS). Laboratory experiments demonstrated that Chip-SIP can detect isotopic enrichment of 0.5 atom % 13 C and 0.1 atom % 15 N, thus permitting experiments with short incubation times and low substrate concentrations. We applied Chip-SIP analysis to a natural estuarine community and quantified amino acid, nucleic acid or fatty acid incorporation by 81 distinct microbial taxa, thus demonstrating that resource partitioning occurs with relatively simple organic substrates. The Chip-SIP approach expands the repertoire of stable isotope-enabled methods available to microbial ecologists and provides a means to test genomics-generated hypotheses about biogeochemical function in any natural environment.

SUMMARY: During one month, paralarvae of common octopus (Octopus vulgaris) were fed 3 different diets: (1) Artemia sp. enriched with Isochrysis galbana (AI) complemented with sand eel (Hyperoplus lanceolatus) flakes (AH); (2) Artemia sp. enriched with crushed marine zooplankton (AZ); and (3) Artemia sp. cultured with Isochrysis galbana and further enriched with the microalga Nannochloropsis sp. (AN). The highest dry weight (1.6179±0.3861 mg) was registered with the AN diet and the best average survival (67.0%) with the AZ diet. Considering the highest dry weight obtained, the moderate high survival and the fact that with this diet it was possible to attain the adult stage, the AN diet was the most appropriate. The reasons for the best result in growth observed with AN are discussed as: (1) the combination of docosahexaenoic acid (DHA) provided by Isochrysis galbana and the high eicosapentaenoic acid (EPA) content present in Nannochloropsis sp.; (2) the fact that the higher protein/lipid ratio of this diet improves the final dry weight of the paralarvae; and (3) the fact that Nannochloropsis sp. could inhibit the harmful microflora growth in the rearing tank. Regarding nutritional aspects, DHA content per se is not the only determinant factor for growth and survival of O. vulgaris paralarvae, but the presence of a high protein/lipid ratio and a high phospholipid content in the diet could possibly explain the better quality and strength of the paralarvae.Keywords: Octopus vulgaris, paralarvae rearing, growth, survival, protein/lipid, DHA, EPA. RESUMEN: Crecimiento y supervivencia de paralarvas de OctOpus vulgaris (Cuvier 1797) alimentadas con tres dietas basadas en artemia complementadas con laminillas de pescado congelado, zooplancton triturado y microalgas marinas. -Paralarvas de pulpo común (Octopus vulgaris) fueron alimentadas, durante un mes, con tres dietas diferentes: (1) Artemia sp. enriquecida con Isochrysis galbana (AI) complementada con laminillas de lanzón (Hyperoplus lanceolatus) congelado (AH); (2) Artemia sp. enriquecida con zooplancton marino congelado (AZ) y (3) Artemia sp. cultivada con Isochrysis galbana y posteriormente enriquecida con la microalga Nannochloropsis sp. (AN). El peso seco más alto (1.6179±0.3861 mg) fue registrado con la dieta AN y la mejor supervivencia media (67.0%) con la dieta AZ. La dieta AN fue la más apropiada teniendo en cuenta el peso seco más alto, la supervivencia moderadamente elevada y el hecho de que con esta dieta fue posible alcanzar el estadio adulto.
