Biological Relevance of RGD‐Integrin Subtype‐Specific Ligands in Cancer

Abstract: Integrins are heterodimeric transmembrane proteins able to connect cells with the micro‐environment. They represent a family of receptors involved in almost all the hallmarks of cancer. Integrins recognizing the Arg‐Gly‐Asp (RGD) peptide in their natural extracellular matrix ligands have been particularly investigated as tumoral therapeutic targets. In the last 30 years, intense research has been dedicated to designing specific RGD‐like ligands able to discriminate selectively the different RGD‐recognizing int… Show more

“…Another recognition system that is evident in multiple systems is that of the integrin family of molecules that recognize specific peptide sequences such as the RGD sequence in fibronectin or other sequences on ECM molecules such as collagen, laminin, and others (reviewed in [51][52][53][54]). Through a combination of different protein chains of family members, a variety of recognition specificities can be achieved.…”

“…Peptides can also disturb the interaction between tumour cells and tumour-associated macrophages (3) or other immune cells ( 4) and as such promote anti-cancer immune responses or attenuate tumourigenic signalling (Thundimadathil, 2012). The author concluded that the use of peptides (not including monoclonal antibodies) will become more common in future cancer therapy because of their chemical versatility and their target specificity to receptors in a tumour, such as the RGD moiety in receptors found in tumour neovasculature (Sani et al, 2021).…”

“…Integrins are heterodimeric transmembrane protein families, and among these integrins, ανβ3 and ανβ5 are overexpressed in endothelial cells of many types of cancers and are emerging as markers of tumoral neo-angiogenesis. Indeed, many investigations are reported in the field of ανβ3 integrin-mediated bioactive RGD motif ( Figure 2 a)-containing peptides [ 30 ].…”
