Background: Endophytes are microorganisms which live within plant tissues without causing any apparent harm to their host. Endophytes have been found in nearly all plant families and can be a source of metabolites that are potential source of antibacterial drugs. Objectives: In this study, we focused on the isolation of bacterial endophytes from three medicinal plants; Zataria multiflora, Achillea willhelmsii and Calendula officinalis L. and screening them for activities against some human bacterial pathogens isolated from nosocomial infections. Methods: In this descriptive study random samples from asymptomatic leaves and branches of three medicinal plants namely: Zataria multiflora, Achillea willhelmsii and Calendula officinalis L. were collected from Chaharmahal Va Bakhtiari province of Iran in spring 2013. For isolation of endophytic bacteria, the disinfected portions of the plants were distributed onto the isolation media. To examine endophytic bacterial contents, the bioassays were conducted with using growing colonies in peptone agar (PA) and yeast extract agar (YEA) media, then inactivated them by chloroform. To assay antibacterial activity of endophytic bacterial culture broth, filter-sterilized supernatants were poured in cylinders on each bacterial plate. Results: Nine discernible bacterial endophytes were isolated in purification from three medicinal plants. Generally, all isolated endophytes showed acceptable effects against indicator bacteria, however endophytes isolated from Zataria multiflora and Achillea willhelmsii were more effective against Staphylococcus aureus than other endophytes. Conclusions: Endophytes of examined medicinal plants could be a good source of antibacterial substances.

Context: For years, natural products from microbes have been used as drugs. Endophytes are the most important fungi that produce many novel metabolites for potential use in pharmacology and agriculture. Objective: The objective of the present study was to explore new endophytes for novel natural products. Materials and methods: An endophyte BAK-I was isolated from the bark of Kigelia africana (Lam.) Beneth (Bignoniaceae). BAK-I was characterized morphologically and on the basis of ITS-5.8S rDNA sequences. BAK-I was fermented to yield an extract, which was evaluated for its anticancer, antimicrobial, and immunomodulatory activities, using MTT, agar well-diffusion, tube dilution method, lymphocyte proliferation, and pro-inflammatory cytokines (TNF-a) (by macrophages) evaluation assays. For lymphocyte proliferation and pro-inflammatory cytokines studies, four concentrations were evaluated 10, 30, 100, and 1000 mg/mL and the experiments were conducted for 72 and 48 h, respectively. Results and discussion: The BAK-I showed pink cottony growth. SEM studies showed smooth fusoid-oblong conidia with a truncated base. Furthermore, ITS-5.8S rDNA sequence showed 99% homology with the Botryosphaeria dothidea strain suggesting that the endophyte is a strain of the genus Botryosphaeria. Less than 50% growth inhibition of SF295, Lung A-549, and THP-1 cancer cell lines after treatment with BAK-I extract suggested that it did not have significant cytotoxic potential, whereas it is bactericidal for Gram-positive pathogens MRSA and VRE with MIC value 200 and 250 mg/mL, respectively. To elucidate its immunomodulation potential, splenocyte proliferation studies showed that BAK-1 suppressed the T cell proliferation by 50%. TNF-a evaluation studies also showed that the extract inhibited TNF-a production in a concentration-dependent manner suggesting that it had immunosuppressive potential. Inhibition at 10 mg/mL was found to be 55% as against 48% using b-methasone. Conclusion:The results suggested that BAK-I extract can be used as a potential immunosuppressive agent.

O metabólito foi analisado contra o parasita Trypanosoma cruzi, o agente causador da doença de Chagas, e mostrou uma atividade tripanocida (IC 50 9,8 µmol L -1 ) similar ao precursor natural (IC 50 3,7 µmol L -1 ).The biotrasformation of the tetrahydrofuran lignan, (-)-grandisin, by the endophitic fungus Phomopsis sp, obtained from Viguiera arenaria, led to the formation of a new compound determined as 3,4-dimethyl-2-(4'-hydroxy-3',5'-dimethoxyphenyl)-5-methoxy-tetrahydrofuran. [10][11][12] Several non-symmetrical 2,5-diaryltetrahydrofuran lignans, shown in Table 1, have been isolated from Piper species (Piperaceae) containing either piperonyl, veratryl or 3,4,5-trimethoxyphenyl substituent at 2 and 5 positions and methyl groups at 3 and 4 positions attached to the tetrahydrofuran ring, as illustrated by (+)-calopiptin P. futokadsura, P. hancei, P. puberculum, P.wallichii), (-)-galbelgin (5, P. attenuatum, P. futokadsura, P. thomsoni, P. wightii), (+)-grandisin (6, P. polysyphorum), (+)-machilin G (7, P. schmidtii, P. wightii), (+)-veraguensin (8, P. cuneifolium, P. futokadsura, P. puberculum) and (-)-zuonin A (9, P. schmidtii). 13 Other bis-tetrahydrofuran lignans have also been described from Achillea lingulata (Asteraceae). 14 In previous reports, Martins et al. 3 described the isolation of (-)-grandisin (6) from Piper solmsianum and Lopes and co-workers 4,15 from Virola surinamensis (Rol.) Warb, which belong to Piperaceae and Myristicaceae, respectively.
