Introduction. Tripartite motif 47 (TRIM47) belongs to a category of the TRIM family. It takes part in cancer tumorigenesis, thus demonstrating important functions across numerous carcinomas. Unfortunately, it is still elusive towards TRIM47 expression, characteristic, and biological function in brain gliomas. Methods. Public database analysis was applied to analyze TRIM47 expression, and quantitative real-time PCR (qRT-PCR) was applied to detect the expression of TRIM47 in 9 paired tissues of glioma. The Cancer Genome Atlas (TCGA) and the Chinese Glioma Genome Atlas (CGGA) databases were applied to evaluate the overall survival (OS). Gene Ontology (GO) term and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways were applied to analyze differentially expressed gene (DEG) functions. In vitro experiments were performed to validate TRIM47-mediated effects on glioma cell proliferation, migration, and invasion. Results. Compared to that in normal tissues, TRIM47 expression was greatly higher in glioma tissues, and its expression level was associated with different grades of glioma. Our data indicated that highly expressed TRIM47 displayed an association with the poor prognosis of glioma patients. Ablating TRIM47 obviously impeded glioma cell invasion and migration. Conclusion. TRIM47 could modulate glioma cell proliferation, invasion, and migration. Highly expressed TRIM47 exhibited a correlation with poor prognosis. All data imply that TRIM47 is a probable biomarker for glioma and has the potentiality to become a newly generated target for glioma treatment.

Expression analysis of cathelicidin and NK‐lysin in Dabry’s sturgeon ( Acipenser dabryanus )

Antimicrobial peptides (AMPs) are a conserved component of the innate immune response in many species. In the present study, the cDNA sequences encoding two AMPs (cathelicidin and NK‐lysin, comprising 1,576 and 606 bp, respectively) were cloned from Dabry's sturgeon (Acipenser dabryanus). Phylogenetic analysis demonstrated that the two AMPs were clustered together with homologous protein sequences from other fish. NK‐lysin was highly expressed during early embryonic development, suggesting maternal transmission. Tissue distribution analysis showed that cathelicidin had the highest expression in the liver and NK‐lysin was most abundantly expressed in the spleen. In response to Poly I:C treatment, the expression of cathelicidin was upregulated at 12 and 24 hr post induction (hpi), but downregulated at 72 hpi. NK‐lysin mRNA expression increased after treatment with Poly I:C, reaching a peak at 24 hpi. Lipopolysaccharide treatment also induced the expression of two antimicrobial peptide genes. Lipopolysaccharide treatment significantly upregulated the expression of cathelicidin at 6, 24, and 48 hpi, and upregulated NK‐lysin expression at 6 and 12 hpi. These results suggested that two AMPs could participate in the immune response induced by poly I:C or LPS stimulation.

TRIM proteins are a group of highly conserved proteins that participate in a variety of biological processes in innate immunity. However, the roles of the Acipenseridae trim genes in the anti‐bacterial immune response remained uncertain. In the present study, the Dabry’s sturgeon (Acipenser dabryanus) TRIM2, TRIM8, and TRIM59 protein sequences were found to be similar to Chicken (Gallus gallus), with identities 92.8%, 89.1%, and 79.4%, respectively. Meanwhile, the TRIM23, TRIM37, and TRIM82 proteins shared 95.8%, 81.7%, and 78.1% similarity with Zebrafish (Danio rerio) homologs, respectively. Sequence alignment indicated that these six adTRIMs contained incomplete domains at their N‐termini, including a RING‐finger domain and one or two B‐boxes domains. The six adtrim genes were constitutively expressed in immune related tissues, such as the head kidney, and were also expressed in non‐immune related tissues, like the blood, brain, muscle, and liver. After challenge with Aeromonas hydrophila, the expression levels of adtrims changed during the early stage of infection. Stimulation by A. hydrophila upregulated the expression levels of adtrim8 and adtrim59 in the spleen and caudal kidney, while the expression of adtrim82 in the spleen and caudal kidney was inhibited at 12 hr post‐infection. A. hydrophila challenge led to significant upregulation of adtrim2 and downregulation of adtrim23 in the spleen. In addition, adtrim37 was only weakly affected by A. hydrophila stimulation. These results suggested that adtrims were related to the immune response to bacterial infection, which provides the fundamental basis for protecting Dabry’s sturgeon against pathogen attack.
