Bioinformatics analysis suggests that COL4A1 may play an important role in gastric carcinoma recurrence

Abstract: Objective Cancer recurrence is a complicated problem for clinicians that contributes to poor prognosis. This study aimed to use advanced gastric carcinoma genes profiles to predict increased risk of cancer recurrence in order to identify patients in need of adjuvant therapy for prognosis improvement. Methods Differentially expressed genes were identified for advanced gastric carcinoma by analyzing the GSE2685 from the Gene Expression Omnibus database (GEO) using R package. The candidate genes were then obtaine… Show more

“…Previous studies have shown that COL4A1 is essential in tumorigenesis, which contributes to the proliferation, migration and colony formation in many cancers ( Huang et al, 2018 ; Jin et al, 2017 ). COL4A1 has been identified as one of the prognostic biomarkers of several diseases, whose high expression level is usually associated with poor overall survival (consistent with our result HR = 1.332) ( Huang et al, 2018 ; Li et al, 2019 ; Sulpice et al, 2013 ). Another marker, COL4A2 , is involved in proliferation and migration of endothelial cells and induces apoptosis ( JingSong et al, 2017 ).…”

“…COL5A2 has been identified as a hub gene in multiple biological analyses of GC, and enrich in EMT-related pathway [44]. In another example, COL4A1 may confer trastuzumab resistance and promote gastric carcinoma recurrence in GC [45,46]. Zhang et al prior studies reported TREM2 is high expression in GC, and significantly associated with clinicopathological characteristics and prognosis [47].…”

“…Cai et al study proved that GPX3 affected migration and invasion through NFкB/Wnt5a/JNK pathway in GC [39]. The key genes COL1A1, COL4A1, COL5A2 belong to the collagen family, which is a constituent of the ECM component of tumors, and is closely related to tumor proliferation, invasion, drug resistance, and prognosis [7,[40][41][42][43][44][45][46]. For example, the high expression of COL1A1 promotes cell proliferation, invasion, and migration in GC [42].…”

“…A previous study reported that mutations in COL4A1 may affect glycine residues in the protein, which is not conducive for its structural and functional stability (13). COL4A1 mutations may cause several diseases, including those pertaining to the cardiovascular, cerebrovascular, renal, gastrointestinal and circulatory systems (14)(15)(16)(17)(18)(19). Studies have indicated that abnormal expression of COL4A1, as a novel oncogene, is closely associated with the occurrence, development and outcome of a variety of cancer types (14,16,20,21).…”
