Bioimaging of Metals and Biomolecules in Mouse Heart by Laser Ablation Inductively Coupled Plasma Mass Spectrometry and Secondary Ion Mass Spectrometry

Abstract: Bioimaging mass spectrometric techniques allow direct mapping of metal and biomolecule distributions with high spatial resolution in biological tissue. In this study laser ablation inductively coupled plasma mass spectrometry (LA-ICPMS) was used for imaging of transition metals (Fe, Cu, Zn, Mn, and Ti), alkali and alkaline-earth metals (Na, K, Mg, and Ca, respectively), and selected nonmetals (such as C, P, and S) in native cryosections of mouse heart. The metal and nonmetal images clearly illustrated the shap… Show more

“…Although the kidneys appear to be Mn limited, the similar bacterial burdens in the kidneys of wild-type and CP-deficient mice suggest that CP does not contribute substantially to Mn sequestration in this organ (14). To address this possibility, IMS was employed to image metal and protein levels in infected organs (14,(61)(62)(63)(64)(65)(66). Imaging LA-ICP-MS was used to examine metal distribution in the kidneys of wild-type C57BL/6 and CP-deficient mice infected with S. aureus.…”

“…LA-ICP MS is an attractive approach because it offers the possibility of not only mapping but also absolutely quantifying these elements over a wide dynamic range [36]. Recent important applications include the monitoring of elementals in various brain disorders including stroke, Parkinson's and Alzheimer's disease, and cancer [37,38].…”

“…Each of these techniques has its strength and weaknesses that in some case can be nicely compensated by a sister technology. Several examples exist in the literature where at least two imaging MS technologies have been employed to investigate the molecular content of tissue sections [38,[43][44][45].…”

“…As a useful attempt, we recently applied LA-ICP-MS technique to determine and quantify the metal distribution in mouse heart. 42 In the future, it will be important to apply these powerful techniques to further investigate the metal distribution in various human diseases.…”
