Biogenesis of the mitochondrial DNA inheritance machinery in the mitochondrial outer membrane of Trypanosoma brucei

Abstract: Mitochondria cannot form de novo but require mechanisms that mediate their inheritance to daughter cells. The parasitic protozoan Trypanosoma brucei has a single mitochondrion with a single-unit genome that is physically connected across the two mitochondrial membranes with the basal body of the flagellum. This connection, termed the tripartite attachment complex (TAC), is essential for the segregation of the replicated mitochondrial genomes prior to cytokinesis. Here we identify a protein complex consisting o… Show more

“…The current model of the TAC contains 13 components four of which (p197, BBA4, Mab22 and TAC65) are localised to the exclusion zone filaments (Gheiratmand et al 2013;Bonhivers et al 2008;Käser et al 2016) . Six components are in the differentiated mitochondrial membranes, four in the outer mitochondrial membrane TAC60, TAC42, TAC40 and pATOM36 (Schnarwiler et al 2014;Käser et al 2016Käser et al , 2017 and two components are likely associated with the inner mitochondrial membrane (p166, AEP1, (Zhao et al 2008;Ochsenreiter and Hajduk 2006) ). TAC102 is a protein of the unilateral filaments and the most proximal component to the kDNA (Trikin et al 2016;Hoffmann, Jakob, and Ochsenreiter 2016) .…”

“…Polyclonal antibodies against TbMic10‐1 (used in a 1:1,000 dilution) and TbMic34 (diluted 1:2,500 for immunoblots) were obtained upon immunization of rabbits with a synthetic peptide (LRKGFRGSTGVSDRESSGS) or recombinant full‐length protein expressed in E. coli respectively. All other antibodies are described in detail (Käser et al , ).…”

“…The mitochondrial protein TAC102 and other TAC components can be partially solubilized by digitonin extractions as shown in earlier studies [30,34,36] . At a concentration of 0.025% digitonin cytoplasmic components can be separated from crude organellar structures containing the mitochondrial organelle.…”

“…The closest interactor of TAC102 is the transmembrane domain containing protein p166 that is localized at the inner mitochondrial membrane [32] . Three outer mitochondrial membrane components of the TAC (TAC40, TAC42 and TAC60, [33,34] ) as well as two…”
