Biofunctionalization of porcine-derived collagen matrices with platelet rich fibrin: influence on angiogenesis in vitro and in vivo

Abstract: Objectives Porcine-derived collagen matrices (CM) can be used for oral tissue regeneration, but sufficient revascularization is crucial. The aim of this study was to analyze the influence of platelet-rich fibrin (PRF) on angiogenesis of different CM in vitro and in vivo. Materials and methods Three different CM (mucoderm®, jason®, collprotect®) were combined with PRF in a plotting process. Growth factor release (VEGF, TGF-β) was measured in vitro via ELISA quantification after 1,4 and 7 days in comparison to P… Show more

“…A vacutainer system and specific sterile plain vacuum tubes with additional silicone within their coating surface were, respectively, used for solid (A-PRF+, Mectron, Carasco, Italy) and liquid PRF (iPRF, Mectron, Carasco, Italy). PRF was directly manufactured with a fixed angle rotor with a radius of 110 mm at 1200 rpm and a relative centrifugal force of 177 g for 8 min (Duo centrifuge, Mectron, Carasco, Italy) following the manufacturer’s instructions [ 7 ].…”

“…In contrast, autologous platelet concentrates such as platelet-rich fibrin (PRF) that are now broadly used in dental and craniomaxillofacial regenerative medicine may potentially overcome this limitation [ 5 ]. Thus far, the pro-angiogenic effect of the PRF has been demonstrated to mainly impact soft tissue regeneration procedures [ 6 , 7 ], but the complex interplay of different cytokines and growth factors leads to an increased proliferation and differentiation of different cell lines, inter alia, osteoblasts [ 8 , 9 ]. In detail, vascular endothelial growth factor (VEGF), tissue growth factor ß (TGFß), and platelet-derived growth factor (PDGF) have been discussed to trigger vasoformative responses [ 10 ].…”

“…These membranes mostly consist of porcine or bovine collagen and are characterized by a good barrier function [3,6]. As a biomaterial, collagen has a number of properties including low immunogenicity, fast vascularization, promotion of wound healing and even bone regeneration [7][8][9][10][11]. It was shown that the material allows sufficient diffusion of nutrients for cellular proliferation and differentiation [12].…”
