Biofilm formation, invasiveness and colicinogeny in locus of enterocyte and effacement negative O113:H21 ShigatoxigenicEscherichia coli

Abstract: The ability to express three additional phenotypes besides Stx production may be a factor influencing the pathogenicity and persistence potential of O113:H21 STEC.

“…Genotypic and phenotypic studies have been performed in O113:H21 STEC strains isolated from food, environment, animal reservoir, and human infections aiming at characterizing the pathogens and the environmental strains [ 6 , 7 , 11 , 12 ]. These studies also searched for molecular markers, such as virulence genes, for: i) discrimination of environmental strains from those associated with human diseases; ii) evaluation of the capacity of animal isolates in causing human diseases.…”

“…Also, strong phenotypic differences in biofilm formation between E. coli strains are commonly observed (56, 70, 71). A lack of correlation between curli/cellulose production and biofilm formation (71, 72) and between cell hydrophobicity and resulting surface attachment behavior (73) has also been described.…”

“…The application of LRT allowed us to also recover and accurately assemble the plasmids of strains 4 and EH41 (Table S1). MLST types, as inferred from the 7-and 15-gene Whittam schemes, grouped the isolates into two major phylogeographical complexes (Table S2) [59,60]: ST-820 strains, a distinct clonal group found in Australia, and ST-223 strains found in the US [55,64] and elsewhere in the world [65][66][67]. However, two US strains from ground beef, 16 and 53, were classified as ST-820, characteristic for Australian strains.…”

“…Multilocus sequence typing (MLST) has identified two major phylogeographical complexes, where ST-223 strains are found around the world, while ST-820 strains are restricted to Australia [59,60]. In particular, infections with Australian ST-820 strains have been associated with severe clinical complications [61][62][63], while ST-223 isolates from the US [55,64] and elsewhere [65][66][67] have been rarely associated with severe human disease, even though they are frequent contaminants of produce and cattle [68][69][70] and can possess virulence traits similar to the clinical O113:H21 strains [59,60]. There is a dearth of knowledge of the intrinsic genomic make-up and Stx-production associated traits of ST-223 and ST-820 complex isolates that might contribute to the disparity in human disease manifestation.…”
