Most of agricultural production in Latin America consists of smallholder farmers who need the development of sustainable technologies, with costs according to their economic condition. Biofertilizers composed of free-living bacteria promote plant’s growth, improve productivity through the strengthening of its roots, and reduce the amount of synthetic fertilizer applied to the crops. The aims of this chapter are to highlight the microorganisms commonly used in agriculture as biofertilizers and the main researches carried out in several countries of Latin America, and to describe the development of an experimental biofertilizer for maize, based on strains of Azospirillum spp. and Pseudomonas fluorescens, in the highlands (Sierra Region) of Ecuador. Seven phylum and 95 genera o microorganism used as biofertilizers or Plant Growth-Promoting Rhizobacteria (PGPR) are summarized, along with the benefits, challenges and future prospect of their use. The effectiveness of the experimental biofertilizer developed in Ecuador was demonstrated through several experiments at the green house and field, in which it was evident the increase of root’s size, the amount of crocket, the percentage of dry matter, and the crops´ yield. The evaluations, accomplished on farmers’ fields showed 30% of increase in yield and 21% of decrease in the cost of production per kilogram; as a consequence of the use of biofertilizer plus 50% of the recommended chemical fertilization, in comparison with standard farming techniques. Farmers can reduce the application of synthetic fertilizers and sustainably increase crop yield through the use of this technology.

In the current context of climate change and water deficit, the selection of native beneficial microorganisms, such as plant growth‐promoting rhizobacteria (PGPR), has become a trend for sustainable agriculture due to their ability to improve plant–bacteria interaction with a minimal adverse effect on the soil microbiota compared to commercial PGPR. Until now, the production of phytohormones like melatonin (MT) by native PGPR and their effect on endogenous MT levels in plants have been poorly studied. MT is a ubiquitous phytohormone that protects plants against biotic and abiotic stress by improving the tolerance of stressed plants. In this work, the production of MT by two native PGPR, Enterobacter 64S1 and Pseudomonas 42P4, was evaluated and both PGPR were applied in Arabidopsis thaliana plants grown under drought conditions to assess the inoculation effects. Parameters such as plant growth, leaf cellular membrane damage, leaf protective compounds, and endogenous MT levels under drought and irrigation conditions were evaluated. The results demonstrated that the native strains Pseudomonas 42P4 and Enterobacter 64S1 produce MT and increase the content of endogenous MT in A. thaliana plants under drought. These native strains improved the tolerance of arabidopsis plants to drought by preventing oxidative and membrane damages and improving plant growth. To the best of our knowledge, this is the first report on MT production by native PGPR and their effects on endogenous MT levels in arabidopsis plants, setting the bases to elucidate the role of native PGPR on water deficit conditions.

In recent years, foliar inoculation has gained acceptance among the available methods to deliver plant beneficial micro‐organisms to crops under field conditions. Colonization efficiency by such micro‐organisms largely depends on their ability to survive when applied on the leaves. In this work, we evaluated the survival and localization of Azospirillum brasilense Az39 (Az39) in excised soybean leaves. Scanning electron microscopy and confocal laser scanning microscopy of a red fluorescent‐transformed variant of Az39 were used to determine bacterial localization, while the most probable number and plate count methods were applied for bacterial quantification. Microscopic observations indicated a decrease in the number of Az39 cells on the leaf surface at 24 h after treatment, whereas midribs and cell–cell junctions of the inner leaf epidermis became highly populated zones. The presence of Az39 inside xylem vessels was corroborated at 6 h after bacterization. Az39 population did not significantly decrease throughout 24 h. We could visualize Az39 cells on the surface and in internal tissues of soybean leaves and recover them through culture methodologies. These results evidence the survival capacity of Az39 on and inside leaves and suggest a previously unnoticed endophytic potential for this well‐known plant growth‐promoting rhizobacteria strain.
