Bioengineering and computational analysis of programmed cell death ligand-1 monoclonal antibody

Abstract: The trans-membrane proteins of the B7 family programmed cell death ligand-1 (PD-L1) and programmed death-1 (PD-1) play important roles in inhibiting immune responses and enhancing self-tolerance via T-cell modulation. Several therapeutic antibodies are used to promote T-cell proliferation by preventing interactions between PD-1/PD-L1. Recombinant technology appears to be quite useful in the production of such potent antibodies. In this study, we constructed recombinant molecules by cloning variable regions of … Show more

“…Currently, more antibodies targeting PD-L1 have also become commercially available (28,29). Novel pharmaceutics development like computational analysis of PD-L1 may be allowed for the accurate determination of antigen-antibody interactions, which could improve the efficiency of immunotherapy (30,31). However, the clinical practice of anti-PD-1 monotherapy only involves some special cancer species, and the efficacy is still controversial (32, 33).…”

“…The scoring function was used to perform rigid docking of the two proteins, and the docking yielded 100 conformations. We selected the protein binding model with the lowest docking energy score and used PyMOL 12.1 to display the 3D graphics of this model 44 …”
