Biodistribution study of 99mTc‐labeled LDL in B16‐melanoma‐bearing mice. Visualization of a preferential uptake by the tumor

Abstract: Since there is strong evidence of a preferential LDL accumulation in tumor cells, LDL might be of interest for tumor imaging. We have tested the ability of 99mTc-LDL in tumor imaging with B16-melanoma-bearing mice as a model for further applications in human studies. The LDL fixation rate was higher with 99mTc-labeled LDL than with 125I labeled LDL. Since technetium-99m remains trapped in the cells, 99mTc-LDL is a well-adapted radioligand because of information given by this radiotracer on the receptor metabol… Show more

“…This is supported by the fact that B16 tumors are known to over-express LDL receptors. 13,26 The ratio of radioactivity above unity observed for all the organs after injection of 111 In-LDL is consistent with the presence of LDL receptors in non-cancerous cells. 24,27 The B16 melanoma showed up clearly on scintigraphy and exhibited marked radioactivity after injection of the 111 In-L-L tracer: B16 cells express the LDL receptor abundantly and the tumor is well vascularized.…”

“…In previous studies, using [1251I]LDL, tumour-liver ratios of 1.5-2 were found (Lombardi et al, 1989;Ponty et al, 1993 (Angelin et al, 1983). The expression of LDL receptors on tumour cells, which lack remnant receptors, will be unaffected by the diet.…”

“…PONTY et al (1993) demonstraram acúmulo de LDL humana marcada com Tc em tumores implantados (melanoma, B16F10) de camundongos (C57bl/6). Após oito dias do implante do tumor, o acúmulo de LDL detectado era maior no tumor do que no fígado ou rim.…”
