Biodegradable Nanoparticles Made of Amino-Acid-Based Ester Polymers: Preparation, Characterization, and In Vitro Biocompatibility Study

Abstract: Abstract:A systematic study of fabricating nanoparticles (NPs) by cost-effective polymer deposition/solvent displacement (nanoprecipitation) method has been carried out. Five amino acid based biodegradable (AABB) ester polymers (four neutral and one cationic), four organic solvents miscible with water, and eight surfactants were tested for the fabrication of the goal NPs. Depending on the nature of the AABB polymers, organic solvents and surfactants, as well as on the fabrication conditions, the size (Mean Par… Show more

“…Different types of fluorescently tagged NPs were prepared according to the polymer deposition/solvent displacement (nanoprecipitation) method under the optimal conditions previously established for the pseudo-proteins-amino acidbased biodegradable polymers. 20 Non-PEGylated NPs (labeled TK-1F and TK-5F NPs) were prepared as follows: 6.0 mg of PEA 8L6 (in case of TK-1F NPs) or the 70/30 mixture of PEAs 8L6/8R6 (ie, 4.2 mg of PEA 8L6 and 1.8 mg of cationic PEA 8R6) (in case of TK-5F NPs) were dissolved in 1.0 mL of DMSO (organic phase) along with the 6.0 mg of FDA (ie, 0.1 wt% from the polymer weight) and dropwise added (dropping rate 12 drops/min) to 10.0 mL of water (inorganic phase) containing 50.0 mg of the surfactant Tween 20 (organic:water phases ratio, 1:10 v/v) at a stirring rate of 700 rpm using a magnetic stirrer. All manipulations were carried out at room temperature.…”

“…The surface and bulk PEGylated NPs (samples TK-4F and TK-8F NPs) were prepared using so-called modified nanoprecipitation method 20 as follows: 6.0 mg of PEA 8L6 (in case of TK-4F NPs) or the 70/30 mixture of PEAs 8L6/ 8R6 (in case of TK-8F NPs) were dissolved in 1.0 mL of DMSO along with the 6.0 mg of FDA. A half portion of the biodegradable surfactant PEG-PEA (25.0 mg) was also dissolved in the organic phase.…”

“…The NPs were prepared according to the polymer deposition/solvent displacement (nanoprecipitation) method under the optimal conditions which were established previously for amino acid based biodegradable ester polymers [37]. All the experiments of the NPs fabrication were performed at r.t.: 6.0 mg of polymer was dissolved in 1.0 mL of the organic solvent miscible with water, DMSO.…”

“…Currently, it is believed that nanofluids are next-generation fluids, which, compared to ordinary liquids, offer greater possibilities, both in terms of dielectric properties and improved heat-exchange efficiency [35]. The area of potential application of nanofluids is very wide and covers many important areas, such as electronics, medicine, and electrical power engineering [36][37][38][39].…”
