Biodegradable Chitosan Nanoparticle Coatings on Titanium for the Delivery of BMP-2

Abstract: A simple method for the functionalization of a common implant material (Ti6Al4V) with biodegradable, drug loaded chitosan-tripolyphosphate (CS-TPP) nanoparticles is developed in order to enhance the osseointegration of endoprostheses after revision operations. The chitosan used has a tailored degree of acetylation which allows for a fast biodegradation by lysozyme. The degradability of chitosan is proven via viscometry. Characteristics and degradation of nanoparticles formed with TPP are analyzed using dynamic… Show more

“…Chitosan nanoparticles have characteristics like pH sensitivity, biocompatibility, and low toxicity, making them a promising candidate for new controlled release drug systems development. An interesting chitosan nanoparticles feature is the possibility to carry hydrophilic drugs such as peptides and proteins due to their hydrophilic character (Piras et al 2015;Poth et al 2015;Hecq et al 2015). Another relevant characteristic is the positively charged chitosan nanoparticle surface, which promotes electrostatic interaction with biological membranes that are negatively charged and improvement in nanoparticle stability in the presence of biological cations (Agnihotri et al 2004;Laranjeira and Fávere 2009).…”

“…The biological activity of an incorporated BMP-2 was observed in an in vivo study in mice. The results showed that biodegradable CS-TPP coatings can be employed to present biologically active BMP-2 on usual implants 54 . A layer-by-layer technique has been established to formulate a stable BMP loaded or collagen/hyaluronic acid (Col/HA) polyelectrolyte multilayer (PEM) film on titanium coating to enhance osseointegration.…”

“…e natural high molecular polymers mainly include heparin [28], chitosan [29], gelatin [30], and albumin [31], while synthetic high molecular polymers are mainly polylactic acid (PLA), poly(lactic-co-glycolic acid) (PLGA), and polycaprolactone (PCL). Nanoparticles can be easily modified to increase their targeting capability.…”
