Bioconversion of quercetin and rutin and the cytotoxicity activities of the transformed products

Abstract: Quercetin and rutin are well-know flavonoids. In spite of this, the comprehension of their metabolism is still incomplete. In this work, the cytotoxic activity of quercetin and rutin and its metabolites produced by metabolism of filamentous fungi was investigated. Flavonoids metabolism was monitored by HPLC and LC-MS. Both flavonoids were extensively metabolized. Quercetin was converted into metabolite methylquercetin (2) and quercetin glucuronide (3) and rutin into metabolite rutin sulphate (5), methylrutin (… Show more

“…Cytotoxic effects of rutin for 48 h incubation in HL-60 leukemic cells were determined by MTT test. The IC 50 values of rutin were about 14 mg/mL and the concentration-dependent cytotoxic effect of rutin was demonstrated [45]. In human leukaemia K562 cell lines (K562 cells), IC 50 of rutin was found to be 400 μg/mL for 24 h and a dose dependant inhibition of cell viability was reported [46].…”

“…Rutin alone did not cause significant DNA damage between the concentrations of 50-400 μg/mL and it decreased H 2 O 2 -induced DNA damage at the concentrations of 100, 200, and 400 μg/mL in K562 cells [46]. Also, at the concentrations of 0.1-200 μg/mL rutin did not induce genotoxicity in HepG2 cells for 2 h incubation [45]. …”

“…In fact, there are various components of fruits and vegetables which may have an antioxidant (anticytotoxic) or a prooxidant (cytotoxic) effect depending on the concentration (4,12). Moreover, flavonoids are extensively metabolized, and their metabolites can be cytotoxic, showing that bioconversion can counteract the anticytotoxic (antioxidant) capacity of flavonoids (3).…”

“…Different strategies can be used for region-selective and stereo-selective incorporation of functional groups in quercetin core. 51 Pharmacokinetic features of quercetin are regulated and determined by types of sugars attached to quercetin. 49 More importantly, it also predicted possible interactions of the biotransformation products with pharmaceutical protein targets.…”
