Biocompatible Polyhydroxyethylaspartamide-based Micelles with Gadolinium for MRI Contrast Agents

Abstract: Biocompatible poly-[N-(2-hydroxyethyl)-d,l-aspartamide]-methoxypoly(ethyleneglycol)-hexadecylamine (PHEA-mPEG-C16) conjugated with 1,4,7,10-tetraazacyclododecan-1,4,7,10-tetraacetic acid-gadolinium (DOTA-Gd) via ethylenediamine (ED) was synthesized as a magnetic resonance imaging (MRI) contrast agent. Amphiphilic PHEA-mPEG-C16-ED-DOTA-Gd forms micelle in aqueous solution. All the synthesized materials were characterized by proton nuclear magnetic resonance (1H NMR). Micelle size and shape were examined by dyna… Show more

“…Amphiphilic copolymers of polyhydroxyethylaspartamide have also been used to form micelles of size 180 nm with Gd contrast agents . In vivo study in normal mice showed phagocytosis of these polymeric constructs resulting in accumulation in liver and henceforth used for liver imaging . Two types of amphiphilic diblock copolymers, PCL‐b‐P(OEGMA‐FA) and PCL‐b‐P(OEGMA‐Gd), consisting of a hydrophobic poly(3‐caprolactone) (PCL) block and a hydrophilic poly(oligo(ethylene glycol) monomethyl ether methacrylate) (POEGMA) block was covalently attached with folic acid (FA) and DOTA‐Gd moieties, respectively, and were mixed to form nanosized (28 nm) polymeric micelles .…”
