Biochemical Evaluation of Agaricus and Pleurotus Strains in Batch Cultures for Production Optimization of Valuable Metabolites

Abstract: The production of various biochemical compounds such as proteins, glucans and glucanases, from the mycelium of four strains of Basidiomycetes species, Agaricus bisporus, Agaricus subrufescens, Pleurotus eryngii and Pleurotus ostreatus, during batch culture in shaking flasks, was studied. Fungi were cultured for 26 days in defined media with glucose as carbon source and were primarily evaluated for their ability to consume glucose and produce mycelial mass and intracellular polysaccharides (IPS). Results showed… Show more

“…In the case of the trials with L. starkeyi , total intra-cellular polysaccharides (IPS, expressed as g/L and % of DCW) were measured based on a modified protocol published by Argyropoulos et al [ 17 ]. Briefly, 0.05 g of DCW was acidified by adding 20 mL HCl (2.5 M).…”

“…Cellular lipids were converted to their respective fatty acid (FA) methyl-esters that were subsequently analyzed in a gas chromatograph (GC-FID) apparatus (Fisons 8060, Markham, ON, Canada), equipped with a Chrompack (São Paulo, Brazil) column (60 m × 0.32 mm) and a flame ionization detector using helium as a carrier gas (2.0 mL/min), according to Giannakis et al [45]. Finally, intra-cellular polysaccharides (IPS, g/L) were quantitatively determined according to the modified protocol adapted by Argyropoulos et al [46].…”

“…The reduction of organic load through cultivation of edible fungi is gaining particular attention [ 5 , 6 , 7 , 8 , 9 , 10 ]. The cultivation of edible and medicinal mushrooms, especially those belonging to the phylum Basidiomycota, has experienced an increasing growth rate in recent years [ 8 , 11 , 12 , 13 , 14 ]. Commercial mushrooms such as shiitake ( Lentinula sp.…”

“…and wood ear mushrooms ( Auricularia sp.) are rich in protein, carbohydrates, fiber, vitamins and minerals, while low in fat content [ 8 , 14 , 15 , 16 ]. Moreover, bioactive secondary metabolites of mushrooms such as phenolic compounds, β-glucans, sterols, etc., possess health-promoting effects to cure inflammation, hypertension, cancer, hyperlipidemia, hypercholesterolemia and other diseases [ 14 , 17 , 18 , 19 ], rendering therefore the cultivation of mushrooms in both liquid and solid-state fermentation configurations one of the pillars of modern industrial, green and red biotechnology [ 5 , 17 , 20 ].…”
