Biochemical Cytology of Trichomonad Flagellates

Abstract: To determine the localization of several enzymes in Tritrichomonas foetus, the axenic KV-1 strain was grown in Diamond's medium with bovine serum, homogenized in 0 .25 M sucrose, and subjected to analytical differential and isopycnic centrifugation . The fractions were assayed for their enzymatic composition and examined electron microscopically . NADH and NADPH dehydrogenases, about 90% of the catalase, and two hydrolases, agalactosidase and manganese-activated ß-galactosidase I are in the nonsedimentable par… Show more

“…For example, oxygen-reducing NADH oxidases are present in the cytosol of Entamoeba histolytica (289), Giardia lamblia (278), Trichomonas vaginalis (281,356,489), and Tritrichomonas foetus (78,345). Again, we designate eukaryotes that have no requirement for O 2 in their core energy metabolism "anaerobes," using the term anaerobic to indicate that the organisms in question do not require free O 2 for survival and multiplication.…”

“…Subcellular, hydrogenosomal membrane and matrix fractions were assayed for the following enzymes: pyruvate : ferredoxin oxidoreductase, hydrogenase, malate dehydrogenase (decarboxylating), acid phosphatase and NADH oxidase to confirm their purity [6,8]. Protein content was measured by using a dye-binding assay kit (Bio-Rad Labs.…”
