Biochemical Characterization and Vaccine Potential of a Heme-Binding Glutathione Transferase from the Adult Hookworm Ancylostoma caninum

Abstract: We report the cloning and expression of Ac-GST-1, a novel glutathione S-transferase from the adult hookworm Ancylostoma caninum, and its possible role in parasite blood feeding and as a vaccine target. The predicted Ac-GST-1 open reading frame contains 207 amino acids (mass, 24 kDa) and exhibited up to 65% amino acid identity with other nematode GSTs. mRNA encoding Ac-GST-1 was detected in adults, eggs, and larval stages, but the protein was detected only in adult hookworm somatic extracts and excretory/secret… Show more

“…Pichia-derived recombinant Na-GST-1, Na-GST-2, and Na-GST-3 were tested in two separate trials for the protection they afforded against N. americanus L3 challenge in an adapted hamster model established at the Institute of Parasitic Diseases, Chinese Center for Disease Control and Prevention (IPD-CCDCP) (66), under an approved protocol as described previously (65,69). Because of limited capacity in the animal facility, which could not hold more than 60 hamsters at a time, Na-GST-1 and Na-GST-2/-3 were tested separately against an adjuvant-only negative control under the same conditions in each trial.…”

“…Our previous studies have identified a nematode-specific class of parasitic GSTs exhibiting a high affinity for heme (59,69). X-ray structural data suggest that heme binding is due to the association of tetrapyrrole compounds with a large hydrophobic binding pocket that forms during homodimerization of two hookworm GST subunits (3).…”

“…11 These findings led to the development of an experimental human hookworm vaccine comprised of Na-APR-1, together with N. americanus glutathione S-transferase 1 (Na-GST-1) a unique hookworm enzyme that functions in heme binding and detoxification. [12][13][14][15] The goal of the vaccine strategy is to elicit neutralizing antibodies to both enzymes which would inhibit the parasites ability to detoxify and eliminate heme as well as utilize host hemoglobin for survival.…”
