Bioassay of prostacyclin and endothelium‐derived relaxing factor (EDRF) from porcine aortic endothelial cells

Abstract: A cascade superfusion technique has been developed for the differential bioassay of prostacyclin and endothelium-derived relaxing factor (EDRF) released from porcine aortic endothelial cells cultured on microcarriers, packed into a column and perfused. 2 Bradykinin (Bk; 20-100 nM) released prostacyclin (9.6 ± 1.5 nm per 106 cells; mean ± s.e.mean, n = 9) and prostaglandin E2 (PGE2; 2.1 ± 0.6 nm per 10' cells) from the column measured by relaxation of strips of bovine coronary artery (BCA) and rabbit mesenteric… Show more

“…Release ofEDRF and nitric oxidefrom endothelial cells EDRF was released by bradykinin (Bk) from porcine aortic endothelial cells cultured on microcarriers and bioassayed either on spiral strips of rabbit thoracic aorta or on platelet aggregation as described previously (Gryglewski et al, 1986a;Radomski et al, 1987). Briefly, between 0.5 and 1.5 ml of microcarriers (0.8-5.0 x 10' cells) in a total volume of 2 ml were stimulated with Bk (1-100 nM).…”

“…EDNO and PGI 2 may act the synergic way [30] as important anti-thrombotic factor in the atrium. Considering that cultured endothelial cells have shown changes in response to agonists as a result of receptor down-regulation [13,14], the proposed methodology can be more useful due to its easy execution, low cost, good reproducibility and effectiveness in mimicking in vivo conditions.…”

“…CHUA, YL ET AL -Adaptation of bioassay to detect endotheliumderived relaxing factors from the canine atrial endocardium Bras Cir Cardiovasc 2009; 24(2): 225-232 regulation of receptors when the cells are cultured and the response of the cultured endothelium to agonists can be altered [13,14]. In the present experiments, a bioassay technique was developed to detect luminal release of endothelium-derived relaxing factors from intact canine atrial endocardium.…”

“…Moro, Darley-Usmar et al examined the role of PN on isolated rabbit aortic strips [60] denuded of endothelium and superfused in a cascade bioassay [61]. The addition of PN (30-300 M) to the superfusate resulted in concentration-dependent relaxations of all three detector strips and these observed relaxations were inhibited by oxyhemoglobin [61].…”
