Bioactive Lipids in MSCs Biology: State of the Art and Role in Inflammation

Abstract: Lipidomics is a lipid-targeted metabolomics approach that aims to the comprehensive analysis of lipids in biological systems in order to highlight the specific functions of lipid species in health and disease. Lipids play pivotal roles as they are major structural components of the cellular membranes and energy storage molecules but also, as most recently shown, they act as functional and regulatory components of intra- and intercellular signaling. Herein, emphasis is given to the recently highlighted roles of… Show more

“…This is in accordance with the theory that foods containing animal fats in the body turn into eicosanoids, in limited quantities. This substance is needed by the body, but if the levels of eicosanoids exceed normal limits it can cause joint disease [17,18].…”

“…Among quantified lipids, SEA and PGE2 showed a relevant uniformity that was therefore validated in a larger ASC-CM cohort. SEA is an endogenous lipid belonging to the N-acylethanolamides family that acts as an anti-inflammatory/immunomodulatory agent through the downregulation of several proinflammatory cytokines [ 37 ]. Conversely, PGE2 exerts a well-known inflammatory action.…”

“…Nevertheless, in the OA context, it can exert either anabolic or catabolic effects on chondrocytes and synoviocytes depending on its concentration [ 38 40 ]. Evidence suggests also that PGE2 may have an immune stimulatory role by facilitating Th1 differentiation and expanding Th17 T cells [ 37 ].…”

“…A study for single-cell analysis to closely monitor the cell signaling and change in morphological phenotypes can yield further insights into the mechanisms and implications of single-cell variation amongst these cells and can help enhance their clinical efficacy. There is growing evidence that lipids play a vital role in MSC functions and are closely associated with the MSCs morphological phenotype 6,7 . Although lipid metabolism plays a pivotal role in MSCs physiopathology, the number of studies at the moment about the lipidome of MSCs is limited and no strong link has been established to understand the direct relationships between single-cell MSC lipidome and its phenotype.…”
