Bioactive fragment of human interleukin-1 b (163-171) modulates the immune response to synthetic peptides of RESA of Plasmodium falciparum

Abstract: In this study a bioactive fragment, residues 163-171 of human interleukin-1beta (hu IL-1beta), that mimics the functions of the whole IL-1 molecule was chemically linked to two peptide sequences of the ring erythrocyte surface antigen (RESA), an asexual blood stage antigen of Plasmodium falciparum. The immunogenicity of different formulations was studied in different haplotypes of inbred mice. The peptide-IL-1beta conjugates delivered in liposomes showed the maximal antibody production. Antigen entrapped in li… Show more

“…IL-1 is known to have diverse immunological functions besides acting as a first costimulatory molecule during T cell activation. IL-1 possess inflammatory and adjuvant property, so we used a bioactive segment of IL-1 (IL-113) to modulate the immunogenic function of an asexual blood stage antigen of P. falciparum (62) as well as modulating the immunogenicity of HIV peptide antigens (63). It was seen that a nine amino acid fragment of ILl could substitute the entire IL-1 molecule in mounting an effective immune response against these candidate antigens.…”

“…Previous studies in our laboratory have established the signi®cant role of immunomodulators like polytuftsin or the bioactive fragment of human IL-1b in conjunction with liposomal delivery of synthetic peptides of ring-infected erythrocyte surface antigen (RESA) for developing eective subunit formulations against malaria [7,17,18,33].…”
