Piperlongumine is an amide alkaloid found in Piperaceae species that shows a broad spectrum of biological properties, including antitumor and antiparasitic activities. Herein, the leishmanicidal effect of piperlongumine and its derivatives produced by a biomimetic model using metalloporphyrins was investigated. The results showed that IC values of piperlongumine in promastigote forms of and were 7.9 and 3.3 µM, respectively. The IC value of piperlongumine in the intracellular amastigote form of was 0.4 µM, with a selectivity index of 25. The piperlongumine biomimetic derivatives, Ma and Mb, also showed leishmanicidal effects. We also carried out an metabolic degradation study showing that Ma is the most stable piperlongumine derivative in rat liver microsome incubations. The results presented here indicate that piperlongumine is a potential leishmanicidal candidate and support the biomimetic approach for development of new antileishmanial derivatives.

With the increased incidence of antibacterial resistance in microorganisms, combining natural products from plants with antibiotics may be considered interesting alternatives for synergy to attain multitarget effects. In this study, the antioxidant activity of the methanol extract of Ziziphus mucronata and its interactions with antibiotics against bacteria of clinical importance were investigated. While its phytochemicals and antioxidant activities were determined by free radical scavenging assays, the antibacterial activities of the extract and its interactions with the antibiotics were determined by macrobroth dilution and the checkerboard methods. From the results, total phenolic content was 29.67 ± 1.90 mg GAE/100 g, total flavonoid content was 8.72 ± 0.08 mg QE/100 g, and total proanthocyanidin content was 1.94 ± 0.00 mg CE/100 g of dry plant material. e inhibition concentration 50% (IC 50 ) of DPPH, BHT, and ascorbic acid was equal to 0.04 ± 0.02 mg/ml, respectively. ose of the ABTS, BHT, and ascorbic acid were equal to 0.02 ± 0.02, 0.04 ± 0.03, and 0.04 ± 0.02 mg/ml, respectively. e checkerboard assay showed that combining the extract with different antibiotics resulted in synergistic (38.75%), indifferent (30%), additive (28.75%), and antagonistic (2.5%) interactions. e interactions between the extract and antibiotics resulting in enhanced antibacterial activities could have resulted from the antioxidant activities of the extract mopping up the ROS generated by the antibiotics or the ability of both extract and antibiotics simultaneously producing reactive oxygen species with deleterious effects resulting in synergistic antibacterial effects.

Resumo Este estudo traz o tratamento taxonômico das espécies de Piperaceae ocorrentes no estado de Alagoas, com o objetivo ampliar o conhecimento sobre a flora alagoana e dar continuidade aos estudos com a família no nordeste brasileiro. Foram registradas 30 espécies de Piperaceae, distribuídas nos gêneros Peperomia (9 spp.) e Piper (21 spp.), das quais oito são pela primeira vez registradas para o estado de Alagoas (3 e 5 spp., respectivamente). Apesar da maioria dos táxons ocorrerem em áreas florestais, alguns podem ser encontrados em áreas de restinga, brejo de altitude, no agreste, cerrado e na caatinga. São apresentadas chave para a identificação dos táxons ocorrentes, descrições para as espécies analisadas, além de comentários sobre distribuição geográfica, habitat, ação biológica, e ilustrações para aquelas pouco conhecidas.
