Chronic hyperglycaemia produces excessive free radicals which could lead to diabetic complications. Hibiscus sabdariffa Linn. is proven to have high antioxidant and antihyperlipidemic properties. The aim of the study was to investigate the effects of H. sabdariffa polyphenol-rich extract (HPE) in the aorta of diabetic rats. Twenty-four male rats were randomly divided into three groups, i.e. non-diabetes mellitus (NDM), diabetes mellitus (DM) and diabetes mellitus with HPE (DM + HPE). HPE at 100 mg/kg or saline as control vehicle were administered daily through oral gavage for 28 days prior to sacrification and the aorta was isolated for further analysis. The glucose level in DM + HPE was significantly lower compared to DM, indicating its potential as an antidiabetic agent. Conversely, catalase (CAT), superoxide dismutase (SOD) activity and the level of glutathione (GSH) in DM + HPE were significantly higher than DM. The level of malondialdehyde (MDA) and protein carbonyl were significantly lower in DM + HPE than DM. Lipid profile analysis demonstrated a significant decrease in triglyceride, total cholesterol and LDL-C in DM + HPE compared to DM, however, HDL-C was significantly increased. Microscopic observation showed a disrupted morphology of the thoracic aorta in diabetic rats. HPE may exert protective effects by acting as an antidiabetic, antihyperlipidemic and antioxidant agent to the thoracic aorta in diabetic rats.

SummaryIn this study we investigated the influence of in vitro-simulated gastrointestinal digestion on the bioaccessibility and antioxidant activity of polyphenols from blueberries (Vaccinium spp.). Total phenolic, anthocyanin, and flavonoid content was determined, and extract and digesta compositions were analysed by HPLC-ESI-MS/MS. The phenolic compounds were relatively stable under a gastric environment, whereas polyphenols and anthocyanins were unstable under an intestinal environment. The bioaccessibility of polyphenol, anthocyanin, and flavonoid was greatly decreased after the intestinal digestion, and the recoveries were only 13.93%, 1.95%, and 15.68% (the IN sample), respectively. Polyphenolic profile alteration occurred during in vitro-simulated gastrointestinal digestion. Changes of phenolic compound antioxidant activity during digestion correlated with polyphenol, flavonoid, and caffeic acid concentrations. Digested extract cellular antioxidant activity was lower than non-digested extract activity (P < 0.05). Polyphenol dose-response correlations with cellular antioxidant activity were observed. These results indicated that in vitro-simulated gastrointestinal digestion significantly impact polyphenols and their antioxidant activity.

BACKGROUND Hibiscus sabdariffa is commonly used in daily life and its extract is applied widely in food and cosmetics. However, it has not been evaluated for its anti‐aging effects. RESULTS Hibiscus sabdariffa calyx aqueous extract (HSCAE) has shown potential collagenase activity suppression effects, together with tyrosinase activity inhibition, and anti‐oxidation as a free radical scavenger. The current investigation demonstrated that HSCAE was not cytotoxic in skin fibroblasts, and it significantly decreased ultraviolet B (UVB)‐induced reactive oxygen species (ROS) on a flow cytometry assay. Moreover, HSCAE reduced matrix metalloproteinase (MMP) expression, increased tissue inhibition of metalloproteinase (TIMP)‐1 level, and enhanced collagen content by inhibiting collagenase activity. It also blocked mRNA and protein expressions of melanin production pathway key factors, including the microphthalmia‐associated transcription factor (MITF), tyrosinase, tyrosinase‐related protein‐1 (TRP‐1), and dopachrome tautomerase‐2 (TRP‐2). CONCLUSION These results demonstrated, for the first time, the potential of HSCAE as a natural antioxidant with the ability to maintain collagen production and to decrease melanin syntheses under UVB radiation, for anti‐aging effects. © 2019 Society of Chemical Industry
