Recent research tried to analyze the conjugation of some chemotherapeutic drugs to the biologically produced gold nanoparticles (GNPs) in one step, without the use of any additional linkers. GNPs was produced using Fusarium oxysporum and their presence was confirmed using spectrophotometer, transmission electron microscope (TEM), X-ray diffraction (XRD) and fourier transform infrared (FTIR) analyses. In order to carry out the conjugation study, capecitabine, tamoxifen, and paclitaxel were added dropwise to the GNPs solution under stirring condition and spectrophotometer, dynamic light scattering (DLS) and FTIR analyses were performed to prove the successful conjugation. Finally, AGS and MCF7 cell lines were used for methyl thiazol tetrazolium (MTT) assay to determine the toxicity of each drug and its conjugated form. Results showed that the spherical and hexagonal GNPs with maximum absorbance peak around 524 nm and average sizes less than 20 nm were produced. FTIR analysis clarified the presence of proteins on the surfaces of the GNPs. After the conjugation process although the FTIR analysis demonstrated that all the drugs were successfully conjugated to GNPs, MTT assay revealed that unlike the paclitaxel conjugated GNPs, capecitabine and tamoxifen conjugates displayed no toxic effects due to their deactivation and low half-lives. Moreover the average size and polydispersity index (PDI) of the GNPs after conjugation with all the three tested drugs increased. In conclusion different types of drugs could conjugate to the GNPs but it is important to employ high stable forms of the drugs in the conjugation procedure.

Abstract Background Biologic nanoparticles can be produced by the active and passive mechanisms but their properties have not been compared to each other and recent research tries to uncover them.Methods Biomass of Fusarium oxysporum was divided into four groups and pretreated in: room temperature (RT) and refrigerator (for preparation the active biomass forms); autoclave and hot air oven instruments (for preparation the inactive biomass forms). Samples were floated in ddH2O and SNPs were produced after addition of 1mmol final concentration of silver nitrate solution. Production of SNPs was confirmed using visible spectrophotometer, transmission electron microscopy (TEM) and X-ray diffraction (XRD) analyses. SNPs were washed and their concentration was determined using ICP measurement. Plate well diffusion method was used for antibacterial activity. MTT and Annexin V-FITC/ propidium iodide assays were used for cytotoxicity and apoptosis induction assays, respectively. Result Results showed that all the four tested samples produced SNPs and had maximum absorbance peaks about 400-450 nm. The produced SNPs had almost same shapes and sizes and XRD results confirmed the presence of SNPs in all the samples. ICP results showed that except the refrigerator incubated sample, the concentration of the SNPs in all the flasks was similar to each other. Due to the differences in the nature of the applied bacterial strains the produced SNPs had some differences in their antibacterial activities. MTT assay results showed that due to lower SNPs concentration of the refrigerator incubated sample they had less toxic effects than the others. Apoptosis induction results revealed that the biologically produced SNPs induced more apoptosis and less necrosis.Conclusion In conclusion, the active and inactive fungal biomass forms produced SNPs that their nature and activity were not altered.

The worldwide bacterial resistance to a wide range of antibiotics originates a global health concern and calls for the development of new antibacterial agents. Over the recent years, nanomaterials-based agents have been proven to be useful for the effective antibacterial applications. Notably, the gold nanoparticles (AuNPs) draw particular attention due to their biological inertness and easy surface functionalization. It is now established that the antibiotic functionalization on the AuNPs surfaces increases the antibacterial efficacy even towards the antibiotic-resistant bacterial cells. Moreover, the antibacterial efficacy can be further enhanced by photothermal therapy using antibiotic conjugated AuNPs. In this review article, we have reviewed the advances in the development of the synthesis methods of antibiotic conjugated AuNPs and gold nanoclusters, and their antibacterial efficacy. We have also discussed the developments in the theoretical understandings behind the interaction of antibiotic molecules with gold surface and its relation to the antibacterial activity. We believe that few parameters including the selection of antibiotic molecules, the method of its attachment to AuNPs, the purification of antibiotic conjugated AuNPs, and the quantification of conjugated antibiotic are crucial and needs to be properly addressed.
