Through years of evolution, biological soft fibrous tissues have developed remarkable functional properties, unique hierarchical architectures, and -most notably, an unparalleled and extremely efficient deformation ability. Whereas the structure-function relationship is well-studied in natural hard materials, soft materials are not getting similar attention, despite their high prevalence in nature. These soft materials are usually constructed as fiber-reinforced composites consisting of diverse structural motifs that result in an overall unique mechanical behavior with large deformations. Biomimetics of their mechanical behavior is currently a significant bioengineering challenge. The unique properties of soft fibrous tissues stem from their structural complexity, which, unfortunately, also hinders our ability to generate adequate synthetic analogs, such that autografts remain the “gold standard” materials for soft-tissue repair and replacement. This review seeks to understand the structural and deformation mechanisms of soft collagenous tissues, with a particular emphasis on tendon and ligaments, the annulus fibrosus (AF) in the intervertebral disc (IVD), skin, and blood vessels. We examined and compared different mechanical and structural motifs in these different tissue types, which are subjected to complex and varied mechanical loads, to isolate the mechanisms of their deformation behavior. Herein, we focused on their composite structure from a perspective of the different building blocks, architecture, crimping patterns, fiber orientation, organization and their structure-function relationship. In the second part of the review, we presented engineered soft composite applications that used these structural motifs to mimic the structural and mechanical behavior of soft fibrous tissues. Moreover, we demonstrated new methodologies and materials that use biomimetic principles as a guide. These novel architectural materials have tailor-designed J-shaped large deformations behavior. Structural motifs in soft composites hold valuable insights that could be exploited to generate the next generation of materials. They actually have a two-fold effect: 1) to get a better understanding of the complex structure-function relationship in a simple material system using reverse biomimetics and 2) to develop new and efficient materials. These materials could revolutionize the future tailor-designed soft composite materials together with various soft-tissue repair and replacement applications that will be mechanically biocompatible with the full range of native tissue behaviors.

Natural blood vessels have complete functions, including elasticity, compliance, and excellent antithrombotic properties because of their mature structure. To replace damaged blood vessels, vascular grafts should perform these functions by simulating the natural vascular structures. Although the structures of natural blood vessels have been thoroughly explored, constructing a small‐diameter vascular graft that matches the mechanical and biological properties of natural blood vessels remains a challenge. We fabricated a hierarchical vascular graft by electrospinning, braiding, and thermally induced phase separation (TIPS) and named it EBT vascular graft (EBTVG) to simulate the structure of natural blood vessels. The internal electrospun structure facilitated the adhesion of endothelial cells, thereby accelerating endothelialization. The intermediate PLGA fabric exhibited excellent mechanical properties, which allowed it to maintain its shape during long‐term transplantation and prevent graft expansion. The external macroporous structure is beneficial for cell growth and infiltration. Blood vessel remodeling aims to combine a structure that promotes tissue regeneration with anti‐inflammatory materials. The results in vitro demonstrated that the EBTVG had matched the mechanical properties, reliable cytocompatibility, and the strongest endothelialization in situ. The results in vitro and replacement of the resected artery in vivo suggested that the EBTVG combined different structural advantages with biomechanical properties and reliable biocompatibility, significantly promoting the stabilization and regeneration of vascular endothelial cells (VECs) and vascular smooth muscle cells (VSMCs), as well as stabilizing the blood microenvironment .This article is protected by copyright. Small‐diameter vascular grafts having biomechanical properties similar to those of native arteries for the treatment of cardiovascular diseases are still elusive. Here, a hybrid extrusion printing and electrospinning technique is presented in which a layer of electrospun nanofibers is deposited over the printed gelatin‐methacryloyl (gelMA) constructs to improve the mechanical performance of gelMA grafts. Various blends of polycaprolactone (PCL) and poly(L‐lactide‐co‐ε‐caprolactone) (PLCL) polymer solutions are used to produce electrospun nanofibers. The variation of gelMA concentration is found to have a negligible role in the overall strength of the graft. It is shown that the burst pressure and tensile strength of the fiber‐reinforced gelMA constructs are comparable to those of native human arteries. Moreover, the compliance of grafts reinforced by 100% PCL and 75/25% PCL/PLCL nanofibers are found to be similar to human muscular arteries and elastic arteries, respectively. The cytocompatibility assessment shows that gelMA presents a bioactive surface for the endothelial cells to survive and grow. Also, PCL/PLCL electrospun nanofibers offer cellular metabolic activity in the same order of magnitude as observed in the control. Therefore, this hybrid technique opens up new possibilities for the fabrication of tubular constructs in tissue engineering.
