This study aimed to investigate acetylcholinesterase (AChE) presence and its enzymatic activity in Toxoplasma gondii (RH strain) tachyzoites and to test a well-known inhibitor of this enzyme. Tachyzoites were obtained from cell culture (sample 1) and peritoneal fluid of experimentally infected mice (sample 2). The protein concentration was determined for each pellet of tachyzoite. In this study, our hypothesis is that T. gondii has the enzyme AChE just like other parasites, and this knowledge might be helpful to develop new chemotherapy strategies to fight toxoplasmosis. The AChE activity was detected in the parasite using 0.025, 0.05, 0.1, 0.2, 0.4, 0.7, and 1.0 mg mL −1 concentrations of protein from tachyzoites. AChE activity has increased progressively according to protein increase, up to a certain point, when it had reduced activity when higher concentrations of protein were tested. The AChE activity of T. gondii was also inhibited with the use of trichlorfon, similar to what occurs with other parasites. Based on these results, we conclude that the enzyme AChE is present in T. gondii tachyzoites. Trichlorfon is able to inhibit the enzyme detected in this study, which might become an option for chemotherapy.

Determinamos factores de riesgo asociados a la seropositividad para anticuerpos anti-Trypanosoma cruzi en 26 poblaciones rurales, 905 viviendas, 2.156 individuos y 333 caninos en el Estado Lara, Venezuela. La seropositividad fue determinada mediante ELISA y MABA. Los datos fueron obtenidos mediante encuestas entomológicas, demográficas y médicas. Los factores de riesgo fueron establecidos mediante regresión logística binaria. La seroprevalencia humana fue de 7,24% y la canina 6,9%. La seropositividad estuvo asociada positivamente al Rhodnius prolixus, la edad, madre con antecedentes de Chagas, consumo de chimó, presencia de mamíferos y aves en la vivienda, desorden en el domicilio, y anexos de bajareque, nidos y cuevas en el peridomicilio. Negativamente con hábitos de consumo de tabaco y alcohol, antecedentes de cáncer y a depósitos en el peridomicilio. En conclusión, la enfermedad de Chagas en el área rural estudiada es un fenómeno remoto transmitida por R. prolixus y vía transplacentaria, asociada a hábitos socioculturales relacionados con la pobreza, a entornos selváticos y antecedentes médicos del huésped.

Trypanosoma evansi is a worldwide distributed hemoparasite with a strong economic impact in veterinary activities. Despite widespread knowledge about the etiology of the disease caused by T. evansi, there are few detailed studies about the metabolism of this parasite. The aim of this study was to determine the presence of Acetylcholinesterase (AChE) in T. evansi through a strategy of subcellular localization and confocal microscopy. The localization of the AChE by differential and isopycnic centrifugation strategy showed that this enzyme has a predominant localization in the glycosome, similar to hexokinase, and it is not present in either the cytosol or the plasma membrane. This study shows novel data that help to understand the non-neuronal role of AChE in the Trypanosomatidae family.
