Binding of Activated α2-Macroglobulin to Its Cell Surface Receptor GRP78 in 1-LN Prostate Cancer Cells Regulates PAK-2-dependent Activation of LIMK

Abstract: Two characteristics of highly malignant cells are their increased motility and secretion of proteinases allowing these cells to penetrate surrounding basement membranes and metastasize. Activation of 21-kDa activated kinases (PAKs) is an important mechanism for increasing cell motility. Recently, we reported that binding of receptor-recognized forms of the proteinase inhibitor ␣ 2 -macroglobulin (␣ 2 M*) to GRP78 on the cell surface of 1-LN human prostate cancer cells induces mitogenic signaling and cellular p… Show more

“…A2m, which activates p21 activated kinase 2, the latter increases cell motility, was downregulated in response to gold-1a, implying that the compound might indirectly reduce malignancy. 28 Moreover, Mgll, which hydrolyzes 2-arachidonoylglycerol, the latter inhibits invasion of cancer cells, 29 was downregulated by gold-1a. Furthermore, genes downstream of hypoxia-inducible factor 1 (HIF-1) (Egln3, 30,31 Aldoc), overexpression of which accompanies the pathogenesis of many tumors, 32 were downregulated in response to gold-1a.…”

“…GRP-78 has been reported to be translocated to the detergent-resistant fractions of the plasma membrane of these cells (5,6). It has also been implicated to function as a receptor for ␣2-macroglobulin and different viruses through lipid rafts (7)(8)(9). Interestingly, GRP-78 has been shown to possess calcium binding properties with reports of calcium binding affinity ranging from 1-2 mol of calcium/mol of GRP-78 to much larger quantities (10).…”

“…Therefore, it appears to play cytoprotective and antiapoptotic functions. Recently, Grp78 expression was found elevated in many tumors and cancer cell lines, including lung (6, 7), breast (8,9), stomach (10,11), prostate (12,13), colon (14), and liver (15,16) cells. However, it is unclear whether and how of this molecule play roles associated with oncogenesis.…”
