Biliary clearance of bromosulfophthalein in anesthetized and freely moving conscious rat

Abstract: The aim of this study was to investigate the effect of anesthesia on the pharmacokinetics of bromosulfophthalein (BSP) with focus on biliary clearance. The plasma concentration profile and biliary clearance of intravenously administered BSP was compared in conscious freely moving bile duct catheterized rats and rats anesthetized with ketamine or Zoletil. The plasma concentration of BSP in conscious rats was similar to that of anesthetized rats, irrespective of the anesthetic used. There was no significant diff… Show more

“…These results suggested that the decreases in Oatp1a1 and Oatp2b1 expression levels at 1 h after intestinal I/R might cause the suppression of liver uptake of BSP, resulting in the high AUC0-90, the low CLtot and CLbile (Table 3). The CLtot and CLbile values in this study are almost consistent with the previous report in other group (28). Moreover, the difference of CLtot between intestinal I/R rats reperfused for 1 h and sham-operated rats was almost same as the difference of CLbile ( Table 3), suggesting that the low CLtot value of BSP in intestinal I/R rats reperfused for 1 h was caused by the suppression of biliary excretion.…”

“…20,21) Pharmacokinetics of Curcumin in EHBR The jugular vein (for drug administration), carotid artery (for blood sampling), and bile duct of each rat were cannulated with polyethylene tubing (Natsume, Tokyo, Japan) under anesthesia at doses of 25 mg/kg and 25 mg/kg for tiletamine and zolazepam, respectively. 22,23) Cannulae were filled with heparinized saline (20 IU/mL) to prevent blood clotting.…”
