Bile acids increase the activity of the epithelial Na+ channel

Abstract: The epithelial Na(+) channel (ENaC) is a key regulator of Na(+) absorption in various epithelia including the distal nephron and the distal colon. ENaC is a constitutively active channel, but its activity is modulated by a number of mechanisms. These include proteolytic activation, ubiquitination and cell surface expression, phosphorylation, intracellular Na(+) concentration, and shear stress. ENaC is related to the bile acid-sensitive ion channel (BASIC), a channel that is expressed in the epithelial cells of… Show more

“…This scenario is in agreement with bile acid-sensitivity of the related ENaC [43]. In Xenopus oocytes, there are two populations of ENaC: one with a high open probability P o and one that is nearly silent [44].…”

“…In Xenopus oocytes, there are two populations of ENaC: one with a high open probability P o and one that is nearly silent [44]. Interestingly, bile acids further increase P o of already active channels but do not activate silent channels [43]. This is in agreement with the idea that the population of ENaC with an energetically unstable closed state (high P o ) senses bile acids and thus membrane alterations whereas the population with a stable closed state (low P o ) does not.…”

“…Data acquisition and solution exchange were controlled using Cellworks 5. Single Channel Patch Clamp Recordings-Single channel recordings from oocytes were performed as described previously (43). Briefly, 24 -48 h prior to experiments, oocytes were injected with WT rBASIC or ⌬N30 rBASIC coding cRNA (0.008 ng).…”

“…Both these genes had a similar trend of gene expression which reduced in WT mice fed a HCD and a further reduction in Hfe -/-mice fed a HCD. Moreover, both these genes have been suppressed by TGF-β1 (243,247,248). Given the differential expression in Hfe -/-mice with the loss of HFE function -a major histocompatibility complex, one can speculate a role for an altered inflammatory response in altered membrane transporters in steatohepatitis.…”
