Bile acid-FXRα pathways regulate male sexual maturation in mice

Abstract: The bile acid receptor Farnesol-X-Receptor alpha (FRXα) is a member of the nuclear receptor superfamily. FRXα is expressed in the interstitial compartment of the adult testes, which contain the Leydig cells. In adult, short term treatment (12 hours) with FRXα agonist inhibits the expression of steroidogenic genes via the induction of the Small heterodimer partner (SHP). However the consequences of FRXα activation on testicular pathophysiology have never been evaluated. We demonstrate here that mice fed a diet … Show more

“…As Fxrα is expressed in different cell types of the testis, we used a full-knockout model (Baptissart et al., 2016). During aging, more Fxrα −/− males remained fertile than wild-type (WT) (Figure 1A).…”

“…Murine spermiogenesis is divided into the following four phases based on sharp nuclear condensation: round spermatids, elongating spermatids, condensing spermatids and condensed spermatids [ 11 ]. For many decades, Leydig cells have been known to produce testosterone to support spermatogenesis [ 12 , 13 ]. Testosterone levels in the adult testis are maintained at a relatively stable and high level (200–800 nM) [ 10 ].The testosterone levels and Leydig cell functions in NMRs remain largely unexplored.…”

“…A remaining question will be to define the endogenous/physiological ligand(s) of FXRα within the testis. Evidence was previously given that, under physiological conditions, there are detectable levels of BA and that testis can synthetized bile acids ( Baptissart et al., 2014 , Baptissart et al., 2016 , Martinot et al., 2017b , Vega et al., 2015 ). In addition, previous reports demonstrate that steroids such as androsterone could be physiological ligands of FXRα ( Wang et al., 2006 ).…”

“…Fxrα −/– mice are described elsewhere ( Baptissart et al., 2016 , Martinot et al., 2017b , Martinot et al., 2017a ). The mice used in this study were housed in a temperature-controlled rooms with 12-hr light/dark cycles.…”
