Bile acid derivatives from a sponge-associated bacterium Psychrobacter sp.

Abstract: In our search for bioactive metabolites from a marine sponge-associated bacterium Psychrobacter sp., a new bile acid derivative (1), which was assumed to be an artifact, were isolated along with six known (2-7) compounds by bioactivity-guided fractionation. Elucidation of the structure of the new compound was done using a combination of NMR ((1)H, (13)C, HMBC, HSQC, and COSY) and MS spectroscopy. Compound 1 exhibited moderate suppressive effects on both NO and IL-6 production at a concentration of 200 microM (… Show more

“…Members of this genus have previously been described to harbour bioactive compounds and dehalogenases2931. The halogenase gene from this isolate has a very high sequence homology to one of the cDNA sequences obtained from the microbial community of C. crambe , indicating that the gene could be detected by culture dependent and independent methods and is expressed within the microbial community, although Psychrobacter sp.…”

“…In contrast, using the same primers, only one of the 107 isolates (isolate 23D8, Psychrobacter sp) was found to possess a putative halogenase gene. By comparison to the NCBI database of microbial proteins and the Conserved Domains Database29, all of these the retrieved sequences were classified as putative tryptophan halogenases.…”

“…IR spectra were acquired using a Bruker Tensor 27 spectrometer. The 1 H-NMR (600 MHz, CDCl 3 containing TMS) and 13 C-NMR (150 MHz, CDCl 3 containing TMS) spectra were obtained using a Bruker AV-600 spectrometer. The J values are given in hertz.…”
