Microplastics (MPs) and brominated flame retardants (BFRs) represent a synergic threat for aquatic environments and organisms' health status, with an additional concern over food quality and food security for species of commercial interest. In this study, the ingestion of MPs, levels of polybrominated diphenyl ethers (PBDEs), and hexabromocyclododecane (HBCDs) were assessed in organisms from two lakes of Central Italy, with the aims of exploring the bioavailability of these pollutants in freshwater environments, the possible translocation of MPs from digestive to edible tissues, and the relationship between MPs ingestion and bioaccumulation of BFRs. The fish Perca fluviatilis, Anguilla anguilla, Carassus auratus, and the crayfish Procambarus clarkii, all species commercialized for human consumption, were caught in Trasimeno Lake; moreover, P. fluviatilis and Rutilus rutilus were also sampled in Piediluco, a lake strongly influenced by industrial and anthropogenic activities, where fishing for commercial purpose is forbidden. With the exception of C. auratus which showed the highest frequency of MPs ingestion (75%), species from Piediluco Lake exhibited a more elevated percentage of organisms positive to MPs ingestion (45%) and higher levels of PBDEs and HBCDs (mean values of 343 and 792 pg/g, respectively, in P. fluviatilis; 445 and 677 pg/g, respectively, in R. rutilus) than Trasimeno species (25% frequency of MPs ingestion, mean values between 6 and 163 pg/g for PBDEs and 5-107 pg/g for HBCDs). Polyester fibers dominated among MP typologies, and a high occurrence of man-made natural fibers was recorded. The number of MPs extracted in the gastrointestinal tracts of fish and soft bodies of crayfishes positive to MPs ingestion ranged between 1 and 2, whereas no MPs were found in fish filets. Given these results, the risk related to human consumption of Trasimeno organisms appears very low, whereas further investigations are required to better elucidate the possible role of MPs pollution in modulating chemical bioaccumulation in edible tissues. This study contributed to assess both environmental quality and food safety, reinforcing the use of bioindicator species for monitoring plans, in accordance with European recommendations.

Farmed crustaceans are an important component in addressing the rising animal protein demand. The present study determined the concentrations of fourteen elements (Ag, As, Be, Cd, Co, Cr, Cu, Fe, Mn, Mo, Ni, Sn, Pb, and Zn) in the edible abdominal muscle of cultured freshwater crayfish species ( Faxonius virilis; Procambarus acutus acutus ) from Missouri. Also, this paper describes the dietary intake and the human health risks from the consumption of crayfish muscle in the adult population. Overall, 172 animals were captured between February 2017 and January 2018 for assessment. Concentrations of metals (Ag, Be, Cd, Cu, Co, Cr, Fe, Mn, Ni, Pb, Sn, Mo, and Zn) and metalloid (As) in the muscle tissue were determined after microwave-assisted acid digestion by ICP - OES. Health indices (EDI/EWI: estimated daily/weekly intakes; THQ: target hazard quotient; TTHQ: total target hazard quotient; ILCR: incremental lifetime cancer risk; and ∑ILCR: cumulative lifetime cancer risk) were calculated and compared to thresholds. Of all samples, the highest concentrations (mg kg −1 wet weight) of metal(loid)s in muscle were Ag (0.11), As (3.15), Be (0.21), Cd (0.11), Co (0.32), Cr (1.22), Cu (107), Fe (23.0), Mn (8.54), Mo (0.62), Ni (2.65), Pb (1.76), Sn (5.91), and Zn (19.2). In both species, the average As, Cd, and Zn concentrations were below the legal limits. However, the levels of Cu, Pb, and As, in some samples, were in exceedance of the maximum levels. In both species, a significant correlation (p < 0.05) was observed between the carapace length (CL) and animal body weight (BW). In P. acutus , CL, BW, and animal total length were homogenous (p > 0.05) among the sexes. Non-parametric Kruskal–Wallis test results indicated significant differences (p < 0.05) in the levels of As, Be, and Zn in F. virilis , and Be and Cr in P. a. acutus among the genders. Significant inter-species differences (p < 0.05) were observed in the levels of Be, Ni, and Pb and the growth factors. The EDI/EWI values were below the permissible limits. THQ and TTHQ values, being below 1.0, indicated no probabilistic health risk. Regarding carcinogenic risk, only As and Ni indicated cancer risk (ILCR >10 −5 and ∑ILCR >10 −5 ) to the adult population. High metals/metalloid exposure from crayfish muscle consumption posed potential health hazards to the adult population.

In this study, the joint toxicological characteristics of reproductive toxicity, neurotoxicity, and endocrine disruption (ED) by organophosphorus flame retardants (OPFRs) were regulated by process control. Molecular docking technology, molecular dynamics (MD), 2D-QSAR model, and density functional theory (DFT) were used to develop a health risk regulation scheme for special population such as pregnant women exposed to OPFRs. It was found that MD simulations confirmed the effectiveness of the recommended complementary food scheme (CFS) for the pregnant women with low health risk. When β-lactoglobulin, α-lactoalbumin, milk fat globule membrane (MFGM) protein, ovalbumin (OVA), ovotransferrin (OVT), vitamin, plant pigment, apple polyphenols, and malic acid were present in the CFS, the joint toxicity of OPFRs in pregnant women were significantly decreased by 2 91.18%. The reproductive toxicity played a dominant role in the joint toxicity and could be reduced by 82.48% under the recommended CFS. There was a competitive relationship between the nutrients in the recommended CFS and OPFRs binding to the joint toxic receptor (JTR). The former could easily occupy the target binding spot of the JTR protein, which reduced or prevented the binding of OPFRs to the JTR. In addition, simulation of OPFRs molecular metabolic pathways in pregnant women under the recommended CFS showed that the binding affinity between OPFRs and six metabolic kinases in pregnant women was significantly decreased (-28.85% --87.54%), indicating that the inhibition effect of OPFRs on normal biochemical reactions in the human body was significantly reduced, which to a certain extent verified the effectiveness of the recommended CFS.
