Beyond Gut Instinct: Metabolic Short-Chain Fatty Acids Moderate the Pathogenesis of Alphaherpesviruses

Abstract: Short-chain fatty acids (SCFA), such as sodium butyrate (SB), sodium propionate (SPr), and sodium acetate (SAc), are metabolic end-products of the fermentation of dietary fibers. They are linked with multiple beneficial effects on the general mammalian health, based on the sophisticated interplay with the host immune response. Equine herpesvirus 1 (EHV1) is a major pathogen, which primarily replicates in the respiratory epithelium, and disseminates through the body via a cell-associated viremia in leukocytes, … Show more

“…SCFA's are known for their role in reducing allergic airway inflammation 30,31 . This anti-inflammatory effect of SCFAs against viral infections was specifically described for the equine herpesvirus 1 (EHV1) where the effect was observed at physiological levels 32 . In our study the inclusion of AuraShield L in the drinking water significantly increased the SCFA (lactate, succinate, formate, acetate, propionate, and butyrate) production which justifies the observed effect on the immune system with positive effects on the viral pathogenicity.…”

“… 19 Treatment of vascular endothelial cells with SCFAs decreased the expression of VCAM-1 and ICAM-1, resulting in reduced adhesion of infected monocytes and virus transfer to the endothelium. 20 Acetate treatment during Influenza infection was effective in reducing secondary bacterial pulmonary infections. 21 Based on this evidence gathered before the SARS-CoV-2 pandemic, many researchers indicated that the reestablishment of SCFAs endogenous production could be useful for the prevention and treatment of COVID-19.…”

“…Recently, SCFA were found to downregulate the expression of ICAM-1 and VCAM-1 adhesion molecules on EC. Pretreatment of EC with SCFA decreased adhesion of EHV-1 infected immune cells to EC and viral transfer ( Poelaert et al, 2019a ).…”
