Diabetic neuropathy is a frequent complication of diabetes. Symptoms include neuropathic pain and sensory alterations—no effective treatments are currently available. This work characterized the therapeutic effect of bergenin in a mouse (C57/BL6) model of streptozotocin-induced painful diabetic neuropathy. Nociceptive thresholds were assessed by the von Frey test. Cytokines, antioxidant genes, and oxidative stress markers were measured in nervous tissues by ELISA, RT-qPCR, and biochemical analyses. Single (3.125–25 mg/kg) or multiple (25 mg/kg; twice a day for 14 days) treatments with bergenin reduced the behavioral signs of diabetic neuropathy in mice. Bergenin reduced both nitric oxide (NO) production in vitro and malondialdehyde (MDA)/nitrite amounts in vivo. These antioxidant properties can be attributed to the modulation of gene expression by the downregulation of inducible nitric oxide synthase (iNOS) and upregulation of glutathione peroxidase and Nrf2 in the nervous system. Bergenin also modulated the pro- and anti-inflammatory cytokines production in neuropathic mice. The long-lasting antinociceptive effect induced by bergenin in neuropathic mice, was associated with a shift of the cytokine balance toward anti-inflammatory predominance and upregulation of antioxidant pathways, favoring the reestablishment of redox and immune homeostasis in the nervous system. These results point to the therapeutic potential of bergenin in the treatment of painful diabetic neuropathy.

Anti-inflammatory properties of bergenin in mice

Inflammation is a protective biological reaction against infection and has undesirable effects. Although bergenin (BG), a C-glycoside derivative of 4-O-methyl gallic acid, has many pharmacological properties, no study has demonstrated the activity of BG in inflammatory mediators, such as histamine and prostaglandin E 2 (PGE 2 ). The present study investigated the anti-inflammatory activity of BG obtained from Peltophorum dubium Taub, in edema models induced by carrageenan and inflammatory mediators in Swiss mice. The assessment of its toxicity was also conducted. The evaluations of this study indicated that the pre-treatment with BG (25 mg/kg) significantly reduced edema induced by carrageenan, compound 48/80, histamine, and PGE 2 (p < 0.05). Also, pre-treatment with BG inhibited the recruitment of leukocytes and neutrophils, reducing adhesion and rolling of these cells, myeloperoxidase (MPO) activity, malondialdehyde (MDA) levels, and vascular permeability. Treatment with BG (2,000 mg/kg) showed no toxic signs in hippocratic screening, weight of animals, water and feed intake, vital organs weight and histopathological evaluation, and hematological and biochemical parameters. In conclusion, these results show that the BG obtained from P. dubium Taub has anti-inflammatory activity against different models of inflammation, reducing neutrophil migration and damage caused by oxidative stress and lipid peroxidation. BG did not present toxic effects in the evaluated parameters.

The Peltophorum clade (Caesalpinioideae, Leguminosae) comprises 16 genera and approximately 105 species with overall pantropical distributions. The genera Peltophorum (Vogel) Benth. (9 spp.) and Delonix Raf. (11 spp.) are notable for having arboreal habits, ornamental uses, and karyotypes of 2n = 26 and 2n = 28, respectively. The present work sought to characterize the patterns of heterochromatin distribution and rDNA sites, as well as the genome sizes of three representatives of the Peltophorum clade. We analyzed P. pterocarpum (DC.) Backer ex K. Heyne, P. dubium (Spreng.) Taub., and D. regia (Bojer ex 121 Hook.) Raf. using double staining with the fluorochromes CMA and DAPI, fluorescent in situ hybridization (FISH) to detect rDNA sites, as well as flow cytometry to determine their DNA contents. The two species of Peltophorum have a chromosome number of 2n = 26, with meta/submetacentric chromosomes measuring on average 2.33 lm. On the other hand, D. regia has a number of 2n = 28, with larger chromosomes. Genome sizes varied from 1C = 0.90 pg in the two Peltophorum species to 1C = 1.55 pg in D. regia. CMA/DAPI staining showed CMA ? /DAPIbands predominantly in the terminal regions of the chromosomes, with six, four, and three pairs being observed in P. pterocarpum, P. dubium, and D. regia, respectively. FISH revealed the presence of a chromosome pair with 5S rDNA sites in each of the two species of Peltophorum, and on two chromosome pairs in D. regia. The numbers of 35S sites varied from 6 to 24, with synteny being observed with 5S rDNA sites in a pair of chromosomes of P. pterocarpum and in two pairs in D. regia. Our data suggest that cytomolecular analysis could be useful tool for differentiating the species within the Peltophorum clade and that comparative analyses with other species could provide new insights into the karyotypic diversification of the subfamily Caesalpinioideae.
