Berberine Inhibits the Metastatic Ability of Prostate Cancer Cells by Suppressing Epithelial-to-Mesenchymal Transition (EMT)-Associated Genes with Predictive and Prognostic Relevance

Abstract: Background: Over 70% of cancer metastasis from prostate cancer develops bone metastases that are not sensitive to hormonal therapy, radiation therapy, or chemotherapy. The epithelial-to-mesenchymal transition (EMT) genetic program is implicated as a significant contributor to prostate cancer progression. As such, targeting the EMT represents an important therapeutic strategy for preventing or treating prostate cancer metastasis. Berberine is a natural alkaloid with significant antitumor activities against many… Show more

“…EMT is an important step in the invasion and metastasis of cancer cells, and a physiological and pathological phenomenon, characterised by the loss of epithelial features and the acquisition of intercellular characteristics (Liu et al 2015). In the process of EMT, the expression of E-cadherin, a membrane protein mediating the tight junction between epithelial cells, is downregulated, and the expression of connexin between mesenchymal cells, such as N-cadherin and vimentin, is upregulated .…”

“…BBR reverses the phenotypic transition and inhibits the migration of the HCoEpiC cells undergoing EMT through regulation of Smad2 and p-Smad3 in the TGF-β pathway Although a large number of studies have illustrated the antagonistic effect of BBR on EMT in various models (S Kim et al, 2018;Liu et al, 2015),. our knowledge about this effect of BBR on EMT in F I G U R E 1 HCoEpiC cells are treated by SW480-derived CM or induction of SW480 in cultured HCoEpiC cells using two kinds of models and intervention of berberine (BBR).…”

“…The strong anti-tumor growth via induction of cell cycle arrest, apoptosis and inhibition of metastasis have been shown in several in vitro and in vivo studies. [6][7][8] Little, however, is known about the anti-cancer activities of berberine on CCA. The present work was aimed to determine the anti-cancer activities and potential molecular mechanisms of berberine on CCA cell lines.…”
