The mechanisms for chiral recognition by cyclodextrins (CDxs) are discussed. Examples of host-guest systems where the 'lock-and-key mechanism' and the 'three-point rule' are applicable are cited and discussed. Most results reported so far suggest that the ability of native CDxs and chemically modified CDxs to discriminate between enantiomers of guests having a central chirality is low in aqueous solutions. Small ⌬⌬G values for enantioselective complexation of CDxs with amino acids seem to be due to unpredictably small changes in the structures of the complexes of the guest enantiomers. Therefore, it is very difficult to prove the participation of hydrogen bonding in chiral recognition through the 'threepoint rule' in these systems. The use of the Coulomb interaction and the coordinate bonds as point interactions is discussed. CDxs might be good hosts to recognize axial chirality or helicity of guests. Hydrogen bonding in water is assumed in the recognition of the helicity of bilirubin. Very large ⌬⌬G values have been reported for the chiral recognition of the binaphthyl derivatives. Such a system seems to be a suitable model system to study the mechanism of chiral recognition by CDxs.

The FAD analogue, Nh-(6-carboxyhexyl)-FAD, carrying a hexanoic acid residue at the N6 position of the adenine moiety was synthesized. A new semi-synthetic oxidase, N6-(6-~arbamoylhexyl)-FAD-~-arnino acid oxidase, was prepared by reacting the succinimido ester of N6-(6-carboxyhexyl)-FAD with apo-Damino-acid oxidase from pig kidney in the presence of benzoate. Reaction conditions and methods have been developed for preparing pure semi-synthetic and fully active N6-(6-carbamoylhexyl)-FAD-~-amino acid oxidase that contains 1 covalently bound FAD analoguehbunit, as verified by redialysis, ultraviolet spectrophotometry, electrospray ionization (ES1)-MS and peptide mapping.Presumably, the Nb-(6-carbamoylhexyl)-FAD moiety of this semi-synthetic D-amino-acid oxidase (DAAO), selectively bound to Lys163, has a structurally similar position to that of the non-covalently bound FAD of the native holoenzyme, since both DAAO forms show very similar kinetic properties (semi-synthetic DAAO, V,,,(app) = 17.7 pmol min-' mg-'; K,(app) = 4.5 mM; native holo-DAAO, V,,,,, = 12.2 pmol min-' mg-'; KM = 1.8 mM). Compared with the native holo-D-amino acid oxidase, this new semi-synthetic N6-(6-carbamoylhexyl)-FAD-~-amino acid oxidase is a considerably more stable enzyme that shows meso-thermostability and withstands inactivation on dilution. Probably, the lack of dissociation of FAD and, consequently, the absence of the instable apoenzyme are responsible for these phenomena. Preliminary investigations resulted in finding convenient and reproducible crystallization conditions for Nh-(6-carbamoylhexyI)-FAD-~-amino acid oxidase. The single crystals, obtained by the sitting-drop method using ammonium sulfate as precipitant, belong to the tetragonal space group I422 with cell dimensions u = 16.3 nm, c = 13.6 nm. The crystals diffract to 0.3-nm resolution, with two molecules being present in the asymmetric unit, demonstrating the two-subunit quarternary structure of this semi-synthetic D-amino-acid oxidase.

Summary1. The uptake of P32 by cervical carcinoma has been studied in 20 patients before radiotherapy, in 6 patients after treatment, and in 6 normal cervices.2. The malignant tissue takes up approximately six times more P32 per gram of tissue than the normal.3. In the treated carcinoma of the cervix, where the tumour has disappeared, the P32 uptake is reduced almost to normal values.4. In two cases of carcinoma of the cervix, where the tumour had shown no response to irradiation, the P32 uptake was as high as that before treatment.5. Comparison of the P32 uptake by malignant tumour and neighbouring tissues, considered as normal, may not be of significance. Although they may appear normal histologically, biological changes in the direction of increased cell activity may have already started.6. It was concluded that P32 uptake may be useful in detecting early malignant change, and for assessing the results of treatment.
