Bepotastine besilate for the treatment of pruritus

Abstract: Bepotastine besilate 1.5% ophthalmic solution is a safe and effective treatment option for allergic conjunctivitis associated pruritus. Side-effect profile is similar to other ocular antihistamine agents. Additional comparative-effectiveness studies would further advance its clinical use. Oral bepotastine is a safe and effective treatment option approved in Japan for allergic rhinitis, urticaria and pruritus associated with skin diseases.

“…These agents provide the benefits of two classes of drugs: the immediate relief of antihistamines with the prophylactic benefit of mast cell stabilizers, and as well some have been shown to have other actions including inhibition of eosinophil migration and other mediators of inflammation (e.g. IL-5, PAF, LTB4) [47]. These are used to ameliorate symptoms but may be augmented by other treatments (e.g.…”

“…Dual action agents group includes drugs such as Olopatadine, Ketotifen, Bepotastine, Alcaftadine and Azelastine which act both as H1 antihistamines and MC stabilizers. Some of these drugs have multiple action and immunological effects, for instance, Bepotastine may have a role in inhibition of eosinophil migration, IL‐5, leukotrienes (e.g, LTB4) and PAF . Additional properties of Alcaftadine include H2 and H4 receptor inverse agonism, inhibition of MC histamine release, decreased eosinophil chemotaxis and activation .…”
