Benzolignanoid and Polyphenols from Origanum Vulgare

Abstract: A new dihydrobenzodioxane derivative, origalignanol (10), together with nine polyphenolic compounds, salvianolic acid A (1), salvianolic acid C (2), lithospermic acid (3), apigenin (8), and luteolin 7-O-b-D-xylopyranoside (9), were isolated from the aqueous ethanolic extract of the aerial parts of Origanum vulgare for the first time. The structure of new compound 10 was determined on the basis of spectroscopic methods. Compound 5 is probably an artifact formed during the isolation. Compounds 1, 2 and 3 showed … Show more

“…Fraction M3-4-17, H 2 O/MeOH (1:1) eluate, was further purified by silica gel cc eluting with n-hexane-Me 2 CO (12:1) to give 4-allyl-2-methoxyphenyl 2-methylbutanoate (4, 1.6 mg). In addition, fraction M3-5 was reseparated by silica gel cc eluting with CH 2 Cl 2 -MeOH (25:1-5:1), followed by Sephadex LH-20 cc (MeOH-H 2 O, 1:1-3:1) repeatedly to give ferulic acid (2.6 mg) (Aoki et al, 1982), p-coumaric acid (11.2 mg) (Lin et al, 1999), 1H-indole-3-carboxylic aid (2.0 mg) (Pouchert and Behnke, 1993b), kaempferol-7-O-b-Dglucopyranoside (3.3 mg), isoquercitrin (13.5 mg) (Agrawal, 1989a), and apigenin-7-O-(6 00 -methyl ester)-glucuronide (2.6 mg) (Lin et al, 2003). Kaempferol (5.7 mg) and quercetin (6.3 mg) were isolated from fraction M5 through Sephadex LH-20 cc (MeOH-H 2 O, 1:1).…”

“…Repeated chromatography was required to obtain highly purified Sal A. The structure of Sal A was identified by spectra (IR, 1D-and 2D-NMR, and MS) analyses and also compared with an authentic sample (Lin et al 2003). The purity of Sal A was determined by reverse-phase high-pressure liquid chromatography (HPLC) with a phenyl column (250 Â 4.6 mm i.d.).…”

“…Purification of the n-BuOH soluble fraction of the crude MeOH extract of the leaves afforded one new compound, Centropogin A (1) and three known compounds (2)(3)(4). The three known compounds were identified as Luteolin-7-O-xyloside (2) [15,16], Luteolin-7-O-[β-apiofuranosyl-(1→2)]β-xylopyranoside (3) [17], and Luteolin-7-O-[β-glucopyranosyl-(1→2)-α-rhamnosyl-(1→6)] β-glucopyranoside (4) [17], by comparison of their spectral data with those published in the literature.…”
