Benzofuran Small Molecules as Potential Inhibitors of Human Protein Kinases. A Review

Abstract: Kinases are known to regulate the majority of human cellular processes such as communication, division, metabolism, survival and apoptosis therefore they can be promising targets in cancer diseases, viral infection and in other disorders. Small molecules acting as selective human protein kinase inhibitors are very attractive pharmacological targets. This review presents a number of examples of biologically active natural and synthetic benzo[b]furans and their derivatives, such as benzo[b]furan-2- and 3-ones, b… Show more

“…For each calculated structure the test of total energy convergence against the k mesh was performed to achieve the total energy accuracy of 0.1 mRy, sucient for analysis of magnetic ordering effects. The calculations were performed for three types of antiferromagnetic structure using the experimental values of lattice parameters taken from [4].…”

“…Benzofuran derivatives tagged with diverse pharmacophoric groups proved to possess multiple pharmacological activities for the past decades 9,10 . Among these pharmacological activities, benzofuran derivatives exhibit significant inhibitory carbonic anhydrase 11,12 , antioxidant 13 , anti-Alzheimer's 14 , anti-inflammatory 15 , antibacterial 15,16 , antitubercular 17 , as well as anticancer activities 18 . Benzofurans were reported to exert their antiproliferative activities through diverse mechanisms of cellular proliferation inhibition including apoptosis induction [19][20][21][22][23] and VEGFR-2 inhibitory action [24][25][26] .…”

“…Organic compounds containing a benzo[ b ]furan nucleus are known to have a range of biological properties Diverse benzo[ b ]furan‐scaffold‐containing substances have shown pharmacological activities, including anticancer and antiangiogenic, antidepressant, anticonvulsant, and anti‐inflammatory activities . Small molecules derived from benzo[ b ]furans have been shown to be active as inhibitors of human protein kinases, which are able to modify other proteins through phosphorylation processes, and are thus directly related to viral infections and cancer . For example, benzo[ b ]furan‐2‐carboxamide A and benzo[ b ]furan‐2‐carbaldehyde B (Figure ) have been reported to have potential as antiproliferative agents against tumor cells through the inhibition of cyclin‐dependent kinases.…”
