Benlysta-Loaded Sodium Alginate Hydrogel and Its Selective Functions in Promoting Skin Cell Growth and Inhibiting Inflammation

Abstract: Benlysta is a new drug approved by the US Food and Drug Administration (US FDA) in 2019 for the treatment of systemic lupus erythematosus. In this study, we loaded the benlysta in the traditional sodium alginate (SA) hydrogel to investigate the potential application of the drug-loaded hydrogel for skin dressing or hypodermic drug. Live/dead staining images and the CCK-8 results showed that the benlysta-loaded hydrogel could promote the growth of human epidermal cells (HaCat), fibroblasts (L929), and endothelia… Show more

“…Considering the importance of hydrogels’ swelling in achieving a good system for drug delivery to the wound site and the fact that the exudation of inflammatory fluid and tissue fluid slows down the healing process, a dressing with a high swelling capacity leads to the absorption of excess secretions and rapid wound healing 58 . Therefore, we evaluated the swelling behavior of Alg-Fib@Nis-EDTA at 25, 37, and 42 °C (Fig.…”

“…Sodium alginate (SA) is a linear anionic polysaccharide polymer [ 75 ]. Because SA contains a large amount of -COO − , SA has obvious pH sensitivity and can quickly form a gel under extremely mild conditions, which can avoid the inactivation of active substances such as sensitive drugs, proteins, cells, and enzymes.…”

“…Because SA contains a large amount of -COO − , SA has obvious pH sensitivity and can quickly form a gel under extremely mild conditions, which can avoid the inactivation of active substances such as sensitive drugs, proteins, cells, and enzymes. Many wound care products take advantage of the structural similarity between alginate and ECM [ 20 , 75 , 76 ]. Chen et al obtained the OSA-DA-PAM hydrogels through the cross-linking of hydrogen bond and dynamic Schiff’s base reaction between dopamine grafting oxidized SA (OSA-DA) and polyacrylamide (PAM) chains.…”

“…Eventually, this benlysta loaded sodium alginate hydrogel will give new ideas for the treatment and management of skin tissue defects. 38 Recently, self-healing hydrogels as a replenishable substance had received considerable interest. Besides, the poor biocompatibility and long self-healing duration of the traditional self-healing hydrogel limited their application in skin tissue regeneration.…”
