Beneficial effects of melittin on ovalbumin-induced atopic dermatitis in mouse

Abstract: Atopic dermatitis (AD) is an inflammatory skin disease characterized by intense pruritus and relapsable eczematous lesions. The hallmarks of AD are defects in the epidermal barrier and immunoglobulin E (IgE)-mediated sensitization to several environmental allergens, as well as an immune disorder mediated by an imbalance toward T-helper-2 response. Melittin, a major component of bee venom, has been studied in various inflammatory diseases. However, the beneficial effects of melittin on mouse with AD-like sympto… Show more

“…BV has an anti-inflammatory effect on AD in this experiment. In previous study, melittin which is a major component of BV also showed an anti-inflammatory function for AD ( 28 ). It seems to be due to melittin, the main component of BV.…”

“…Induction of AD was performed according to the method given by Kim et al ( 28 ) Briefly, All the mice, with the exception of those in the NC group, were intraperitoneally inoculated mixed with 10 µg of chicken OVA (grade V) (Sigma-Aldrich; Merck KGaA, Darmstadt, Germany) mixed with 4 mg of aluminum hydroxide (ImjectAlum ® ; Thermo Fisher Scientific, Inc., Waltham, MA, USA) in a volume of 200 µl three time at intervals of one-week intervals (i.e., on days 0, 7, and 14). They were then anesthetized by isoflurane inhalation (Ifran ® ; HANA Pharm, Seoul, Korea).…”

“…Western blotting was performed as described previously. 31 Primary antibodies used in the study were as follows: anti-E-cadherin, vimentin (BD Biosciences), anti-VE-cadherin, TGF-β1, α-SMA, TNF-α, IL-6, S100A4, Fibronectin, collagen Ⅰ (Abcam), antip-Smad2/3, t-Smad2/3, Slug, Snail, t-ERK1/2, p-ERK1/2, YAP/TAZ, p-Akt, p-JNK (cell signaling), anti-Smad4, IL-1β, and TIMP-1 (Santa Cruz Biotechnology). β-actin (Sigma-Aldrich) was used to control the protein loading.…”

“…Some research reported that the ovalbumin has molecular weight 43 kDa (Fish et al 1969), 45 kDa (Chun et al 2011) or ranging from 43 to 45 kDa (Urisu 2001). This protein has an immunogenic property and used by scientist to induced allergic asthma (Alenius et al 2017) and atopic dermatitis (AD) on an animal model (Kim et al 2017). Ovalbumin protein is containing 385 amino acid residues, which is on 323-339 amino acid residue are have IgEbinding activity and promote histamine release (Johnsen and Elsayed 1990).…”
