Beneficial effects of Cuscuta chinensis extract on glucocorticoid-induced osteoporosis through modulation of RANKL/OPG signals

Abstract: Cuscuta chinensis Lam. (Convolvulaceae) is an important herbal medicine widely used to improve sexual function, treat osteoporosis, and prevent aging, and has been reported to exhibit anti-osteoporotic effects in vitro. However, the activity of Cuscuta chinensis Lam. on glucocorticoid-induced osteoporosis still remains unclear. The present study aimed to assess the protective effect and the underlying mechanism of action of Cuscuta chinensis extract (CCE) against glucocorticoidinduced osteoporosis in vivo. Spr… Show more

“…Our findings indicate that mEVs positively act on the RANKL/OPG system by decreasing their ratio systemically and locally. These data corroborate with other studies, in which a decrease in this ratio is associated with reduced osteoclastogenesis after treatment with plant and root extracts in models of bone loss, such as ovariectomy (Sun et al, 2019;Zhu et al, 2020) or glucocorticoid (Mo et al, 2019). An amelioration in bone structure with the treatment of mEVs was shown by improving the bone stiffness despite some slight effect in the femur microarchitecture in OVX mice.…”

“…RANKL activates osteoclast differentiation, thus promoting bone resorption by competing with OPG to bind to receptor RANK. Overexpression of RANKL can lead to excessive activation of osteoclasts, which can lead to osteoporosis ( 41 ). OPG was also a member of the TNF superfamily.…”
