Chemical Fractions and Availability of Zinc in Winter Wheat Soil in Response to Nitrogen and Zinc Combinations

Nitrogen (N) is critical for zinc (Zn) accumulation in winter wheat grain via enhancing Zn absorption into plant roots. This paper explored a possible mechanism for enhanced absorption of Zn in winter wheat by N combined with Zn application based on the Zn bio-availability in soil. A pot experiment with three N application rates (0.05, 0.2, and 0.4 g kg-1), two Zn application rates (0 and 10 mg kg-1), without and with plants was conducted. The results showed that high N (N0.2 and N0.4) combined with Zn (Zn10) application significantly increased the yield, yield components and Zn and N concentrations in winter wheat shoots and grain. The available Zn concentration in soil with and without plants was increased by N0.2Zn10 and N0.4Zn10 treatment at each growth stage. N0.2Zn10 and N0.4Zn10 treatment significantly decreased the pH in soil without plants but had different influences on the pH in soil with plants, which depended on the different N application rates and growth stages. Meanwhile, N0.2Zn10 and N0.4Zn10 treatment decreased the exchangeable Zn but increased loose organic-, carbonate- and Fe-Mn oxides-bound Zn concentrations in soil without plants. The exchangeable, loose organic- and carbonate-bound Zn concentrations in soil with plants was increased by N0.2Zn10 and N0.4Zn10 treatment at different growth stages. Different rates of N combined with Zn application influenced the proportion of Zn in different fractions in soil with and without plants at different growth stages. At Zn10, N0.4 treatment showed higher yield, N and Zn concentrations in shoot and grain, and available Zn concentration in soil, but lower pH in soil than N0.2 treatment. In addition, soil without plants had higher available Zn concentrations and lower pH than did the soil with plants. There were significant differences in Zn chemical fractions concentrations and proportions between the soils with and without plants at each growth stage. Therefore, combined influence of roots and the combination of N and Zn (especially N0.4Zn10 treatment) improved the bio-availability of Zn in soil via changing the soil pH and promoting the transformation and distribution of Zn in different fractions.

Background: Root-microbiome interactions are of central importance for plant performance and yield. A distinctive feature of legumes in this context is that they engage in symbiosis with rhizobia, which are abundant in soils and include both symbiotic and non-symbiotic bacterial strains. If and how the capacity of rhizobia to form symbiosis modulates root-associated microbiomes are not well understood. Results: We address this question by inoculating soybean (Glycine max) plants with wild type (WT) or a noeI mutant of Bradyrhizobium diazoefficiens. The noeI mutant produces a defective Nod factor and is thus compromised in its ability to establish functional symbiosis. Compared to soybean plants inoculated with WT rhizobia, plants inoculated with the noeI mutant showed a significant decrease in nodulation and root-flavonoid exudation, and exhibited strong changes in microbiome assembly in the rhizosphere and the rhizoplane. NoeI mutant-inoculated roots exhibited reduced diversity, co-occurrence interactions and a substantial depletion of beneficial microbes on the roots. The effects of the noeI mutation were absent in soils without plants, demonstrating that they are plant dependent. Complementation experiments showed that flavonoid supplementation is sufficient to restore recruitment of beneficial microbes. Conclusion: The results illustrate that the capacity of a rhizobium to form microbial symbiosis dramatically alters root-associated microbiomes, most likely by changing root exudation patterns. The results of this study have important implications for our understanding of the evolution of plant-microbiome interactions in the context of plant-bacterial symbioses.

River discharge is of great significance in the development of water resources and ecological protection. There are several large ungauged catchments around the word still lacking sufficient hydrological data. Obtaining accurate hydrological information from these areas is an important scientific issue. New data and methods must be used to address this issue. In this study, a new method that couples unmanned aerial vehicle (UAV) data with the classical slope–area method is developed to calculate river discharges in typical ungauged catchments. UAV data is used to obtain topographic information of the river channels. In situ experiments are carried out to validate the river data. Based on slope–area method, namely the Manning–Strickler formula (M–S), Saint-Venant system of equivalence (which has two definitions, S-V-1 and S-V-2), and the Darcy–Weisbach equivalence (D–W) are used to estimate river discharge in ten sections of the Tibet Plateau and Dzungaria Basin. Results show that the overall qualification rate of the calculated discharge is 70% and the average Nash–Sutcliffe efficiency coefficient is 0.97, indicating strong practical application in the study area. When the discharge is less than 10 m3⁄s, D–W is the most appropriate method; M–S and S-V-1 are better than other methods when the discharge is between 10 m3⁄s and 50 m3⁄s. However, if the discharge is greater than 50 m3⁄s, S-V-2 provides the most accurate results. Furthermore, we found that hydraulic radius is an important parameter in the slope–area method. This study offers a quick and convenient solution to extract hydrological information in ungauged catchments.
