BDNF induces transport of PSD-95 to dendrites through PI3K-AKT signaling after NMDA receptor activation

Abstract: The N-methyl-D-aspartate receptor (NMDAR), brain-derived neurotrophic factor (BDNF), postsynaptic density protein 95 (PSD-95) and phosphatidylinositol 3-kinase (PI3K) have all been implicated in long-term potentiation. Here we show that these molecules are involved in a single pathway for synaptic potentiation. In visual cortical neurons in young rodents, the neurotrophin receptor TrkB is associated with PSD-95. When BDNF is applied to cultured visual cortical neurons, PSD-95-labeled synaptic puncta enlarge, a… Show more

“…One correlate of the activation of these pathways is increased synaptogenesis and levels of synaptic PSD95 (27,56). These results are consistent with previous findings of reduced PSD95 transcription and protein expression as well as fewer excitatory synapses and spines in Mecp2 mutant mice (5,11,57,58).…”

“…BDNF belongs to the neurotrophin family and has been shown to play an important role in learning and memory (34,35). BDNF signaling through its primary receptor TrkB has also been shown to play a significant role in downstream effects underlying learning (36)(37)(38)(39)(40). Previous data from our laboratory have shown increased BDNF transcription and translation in the OB following olfactory fear conditioning (41).…”

“…(1-3)IGF-1 treatment phosphorylates the IGF-1 receptor and also leads to activation of the MAPK cascade (data not shown), another signaling pathway that is stimulated by BDNF and is involved in synaptic stabilization (17,46). Although IGF-1 treatment does not increase BDNF levels in MeCP2 mutant mice (as measured with ELISA on whole brain extracts of MeCP2 -/y adult mice, data not shown), it is plausible that it would partially mimic BDNF action by stimulating common downstream signals (23,46). IGF-1 infusion into the brain up-regulates intracortical inhibition and the expression of extracellular matrix molecules (43), which are additional mechanisms that promote the maturation and stabilization of cortical circuits (47).…”
