BD PuraMatrix peptide hydrogel as a culture system for human fetal Schwann cells in spinal cord regeneration

Abstract: BD PuraMatrix peptide hydrogel, a three-dimensional cell culture model of nanofiber scaffold derived from the self-assembling peptide RADA16, has been applied to regenerative tissue repair in order to develop novel nanomedicine systems. In this study with PuraMatrix, self-assembling nanofiber scaffold (SAPNS) and Schwann cells (SCs) were isolated from human fetal sciatic nerves, cultured within SAPNS, and then transplanted into the spinal cord after injury (SCI) in rats. First, the peptide nanofiber scaffold w… Show more

“…PuraMatrix (BD), a self-assembling peptide hydrogel, is a three-dimensional cell culture system for various types of cells and has been applied to regenerative tissue repair. Transplantation of SCs cultured in PuraMatrix promotes their survival in the injured spinal cord and reduces astrogliosis and locomotor impairment (Moradi et al, 2012). Alginic acid sodium hydrogel co-transplantation with SCs reduces their apoptosis when they are placed within the injured spinal cord and enhances SC-mediated recovery of locomotor function after SCI .…”

“…Previous studies suggest that the use of matrices to suspend SCs for transplantation can improve their longterm survival within the injured spinal cord (Patel et al, 2010;Moradi et al, 2012;Wang et al, 2012). Matrigel (BD), a matrix composed of laminin, collagen type IV, heparan sulfate proteoglycans, entactin, and growth factors, has been employed with SC grafting in various models of SCI.…”
