BCLAF1 and its splicing regulator SRSF10 regulate the tumorigenic potential of colon cancer cells

Abstract: Bcl-2-associated transcription factor 1 (BCLAF1) is known to be involved in multiple biological processes. Although several splice variants of BCLAF1 have been identified, little is known about how BCLAF1 splicing is regulated or the contribution of alternative splicing to its developmental functions. Here we find that inclusion of alternative exon5a was significantly increased in colorectal cancer (CRC) samples. Knockdown of the BCLAF1 protein isoform resulting from exon5a inclusion inhibited growth and that … Show more

“…Since SRSF2 is known to function as a splicing regulator, we next decided to take advantage of an RNA-Seq platform to determine the role of SRSF2 in regulating AS of endogenous liver transcripts. We had used this approach with other cells to characterize the biological significance of SRSF10 as a sequencespecific splicing regulator in vivo (18,20,21 Table S2 in the supplemental material). From a total number of 6,133 splicing events, we identified 390 AS events that changed significantly in the HKO samples (Fig.…”

“…SRSF1, SRSF2, or SRSF10 cDNA was cloned into the pcDNA3.0-hemagglutinin (HA) vector. Cells were transfected with small interfering RNA (siRNA) oligonucleotides using Lipofectamine RNAiMAX, while plasmid transfection was performed by using Lipofectamine 2000 (Invitrogen, Carlsbad, CA) as previously described (18,19). Sequences of siRNA oligonucleotides are listed in Table S1 in the supplemental material.…”

“…In addition, Bcl-2 family members exhibit both pro- and anti-apoptotic activities on modulating the intrinsic apoptosis pathway [39]. The balance between Bcl-2 family members is meticulously determined by cell survival or apoptosis [40]. Moreover, impaired splicing profiles of Bcl-2 family members result in differential or opposite effects in regulating cell viability [27].…”

“…24 Although most of studies suggest that Bclaf1 is a tumor suppressor, a recent report indicated that Bclaf1 was oncogenic in colorectal cancers. 25 Thus, the function of Bclaf1 in carcinogenesis is controversial.…”
